酶响应海藻酸钠/阿拉伯木聚糖-丁酸梭菌双重微胶囊体外耐受性分析
In vitro tolerance analysis of enzyme-responsivesodium alginate/arabinoxylan Clostridium butyricum dual microcapsules

Abstract
A dual microcapsule system for Clostridium butyricum(C.butyricum)was developed through encapsulation of sodium alginate(SA)with arabinoxylan(AX),which was extracted from discarded rice bran.Such microcapsules could be coated with protamine sulfate(PRM)on their surface to enhance the environmental tolerance for C.butyricum during the targeted delivery.Our research showed the extraction of rice bran afforded two types of AX,which were mainly composed of xylose and arabinose.The total sugar contents turned out to be quite high in both oligosaccharides,reaching(81.86±0.67)%and(76.87±0.12)%,respectively.Further studies revealed that both types of AX could react with laccase to form dense reticular gel.In addition,they could serve as prebiotics for the utilization of C.butyricum,giving fermented products including lactic acid,acetic acid and butyric acid.Based on the above properties,(SA-AX)@PRM dual microcapsules were prepared using piercing-solidifying method by combination with electrostatic self-assembly,then evaluated by treatment with simulated gastric fluid and high bile salt.Compared with free bacterial cells,C.butyricum embedded in(SA-AX)@PRM system displayed higher survival rates under both treatment conditions,providing 32.4%and 16.2%increase,respectively.Finally,a simulated gastrointestinal tract release experiment was conducted to mimic the targeted release of probiotics in the colon.C.butyricum embedded in(SA-AX)@PRM not only demonstrated good stability in the gastric fluid,but also showed the desired release by pancreatic protease upon the transfer to simulated intestinal fluid.关键词
