基于非靶向代谢组学探讨不同体型成年犬的代谢特征及差异
Study on the metabolic characteristics and differences of adult canines of different sizes based on untargeted metabolomics

Abstract
Objective:To explore the metabolic characteristics and differences among canines with different body sizes,and then to provide better health guidance and disease prevention for canines with different body sizes.Methods:By collecting fresh serum and plasma samples from canines with different body sizes,some blood samples for routine blood analysis,and some samples for blood biochemical and untargeted metabolomics tests to explore the metabolic characteristics of canines with different body sizes.To develop a robust model utilizing partial least squares discriminant analysis(PLS-DA)and employ variable importance in projection(VIP)scores as a means of identifying significant differential.Results:There are significant discrepancies in key serum biochemical markers,including aspartate aminotransferase(AST),total bilirubin(TBil),direct bilirubin(DBil),indirect bilirubin(IBil),and high-density,lipoprotein choles-terol(HDL-C).By using plasma untargeted metabolomics analysis,it identified the canine plasma 1 277 metabolites and divided into 9 categories,lipids and lipid-like molecules,organic acids and derivatives,organic heterocyclic compound,benzoic acids,organic oxides,phenylpropane and polyketones,organic nitrogen compounds,nucleosides and nucleo-tides and analogues,alkaloids and derivatives.Bioinformatics analysis indicated that differential metabolites were en-riched in different KEGG pathways including pyrimidine metabolism,tyrosine metabolism,aminoacyl-tRNA biosynthe-sis,secondary bile acid biosynthesis,as well as arginine and proline metabolism.Among them,the aminoacyl-tRNA biosynthesis pathway was the most significant and wasupregulated.Therefore,the significant differential metabolites L-phenylalanine and L-lysine in this pathway can be used as important targets to distinguish metabolic differences in canine size and body size.Conclusion:This study reveals the metabolic differences between canines with different body sizes,providing more accurate information on the metabolic characteristics of canines with different body sizes.These data can also help us further understand the metabolic characteristics of canines with different body sizes and contribute to more scientific breeding and management.关键词
