摘要

Abstract

[Objective]The persistent sepals of'Yuluxiangli'pear fruits significantly reduces its commercial value.This study aimed to screen and identify key genes involved in sepal development to provide reference genes for molecular breeding of pear fruits with sepal abscission at the molecular level.[Method]'Yuluxiangli'cultivated in vertical trellis was used as tested mate-rial.Based on previous BSA gene mapping and sequencing data,the expression characteristics of candidate genes were verified,and two auxin-amido synthetase genes,GH3 family,were cloned and analyzed for their bioinformatics characteristics.[Result]The genes PbGH3.3(Pbr007550)and PbGH3.4(Pbr030571)ranked second in expression level in the sepals.The CDS lengths of PbGH3.3 and PbGH3.4 were both 1806 bp,encoding 601 amino acids.There were seven amino acid sequence dif-ferences between them,and the encoded protein PbGH3.3 and PbGH3.4 showed the highest homology with MdGH3.1 in ap-ple.They were unstable proteins without signal peptides and were located in the cytoplasm;Analysis using the CCD tool on the NCBI website revealed that they both possessed the conserved domain GH3 superfamily of GH3 protein family.Protein ter-tiary structure modeling showed that they were GH3.1 Auxin amido synthetase involved in auxin homeostasis,belonging to the GH3 protein family.Functional enrichment analysis of GO and KEGG pathways of STRING protein interaction network mem-bers indicated that PbGH3.3 is involved in the plant hormones,especially auxin signaling pathway.[Conclusion]It was sug-gested that PbGH3.3 and PbGH3.4 might play a biological roles in the regulating of sepal in'Yuluxiangli'fruits through the IAA signaling pathway.
