为解析萨湖杂交羊较湖羊肌肉和生长性能提升的关键基因和调控通路,本研究对出生2日龄和8月龄的萨福克和湖羊的杂种F1代(SH)和纯繁湖羊(H)的体重、体高、体长、胸围和管围等生长数据进行比较分析.选取2日龄和8月龄绵羊每组各3只(共12只),利用RNA-seq对背最长肌进行转录组测序,并对测序数据进行生物信息学分析.随机选取10个差异表达基因,通过qRT-PCR对其表达量进行验证.结果表明:1)出生2日龄和8个月的SH的体重、胸围、管围均极显著高于H(P<0.001).2)在2日龄和8月龄SH与H中分别筛选得到684和248个基因显著差异表达,包括与骨骼肌生长发育相关的CDH1、GSTA1-1、GPX2、CDH17和MX2等基因.3)GO和KEGG分析结果显示2日龄SH和H差异表达基因主要富集在控制肌肉生长发育的细胞分化、核小体组织、肌动蛋白丝调控等类别中,8月龄SH和H差异表达基因主要富集在细胞-细胞粘附、谷胱甘肽代谢和PPAR信号通路等类别中.4)qRT-PCR试验结果与转录组测序相一致.综上,萨湖杂交羊在生长性能方面优于湖羊,CDH1、GSTA1-1和GPX2等基因通过G蛋白偶联受体、细胞-细胞粘附、谷胱甘肽代谢通路在萨湖杂交羊肌肉发育过程中发挥重要作用,本研究为探究调控杂交绵羊生长性能的关键基因和功能通路提供参考.

To analyze the key genes and regulatory pathways involved in the muscle and growth performance enhancement of Sahu crossbred sheep compared to Hu sheep,in the study,the growth data of body weight,body height,body length,chest circumference and cannon circumference of F,generation of Suffolk and Hu sheep hybrids(SH)and Hu sheep(H)at 2 days and 8 months of age were compared and analyzed.Three sheep of 2-day-old and 8-month-old were randomly selected from each group(12 sheep in total).RNA-seq was used to sequence the transcriptome of longissimus dorsi muscle,and bioinformatics analysis was performed on the sequencing data.Ten differentially expressed genes were randomly selected and their expression levels were verified by qRT-PCR.The results showed that:1)The body weight,chest circumference and cannon circumference of Sahu hybrid sheep at 2 days and 8 months were significantly higher than those of Hu sheep(P<0.001).2)A total of 684 and 248 genes with significant differential expression were screened in 2-day-old and 8-month-old SH and H,respectively,including CDH1,GSTA1-1,GPX2,CDH17,MX2 and other genes related to skeletal muscle growth and development.3)GO and KEGG analysis showed that the differentially expressed genes were mainly enriched in the categories of cell differentiation,nucleosome organization and actin filament regulation controlling muscle growth and development in 2-day-old SH and H,while the differentially expressed genes were mainly enriched in the categories of cell-cell adhesion,glutathione metabolism and PPAR signaling pathway in 8-month-old SH and H.4)The results of qRT-PCR were consistent with the transcriptome sequencing.In summary,the growth performance of Sahu hybrid sheep is better than that of Hu sheep.CDH1,GSTA1-1 and GPX2 genes play important roles in muscle development of Sahu crossbred sheep through G protein-coupled receptors,cell-cell adhesion,and glutathione metabolic pathways.The study provides a reference for exploring the key genes and functional pathways that regulate the growth performance of hybrid sheep.
