固始鸡胸肌代谢组特征评价及相关基因鉴定OA北大核心CSTPCD

Evaluation of metabolome characteristics and identification of related genes in Gushi chicken breast muscle

[目的]揭示固始鸡优良肉质性状特征与潜在调控机制.[方法]采用液相色谱质谱联用(LC-MS)技术,以45日龄AA肉鸡为对照组,对120日龄固始鸡胸肌代谢物进行了表征,并结合胸肌转录组数据鉴定了差异代谢物(differential accumulated metabolite,DAMs)相关基因.[结果]代谢组分析显示,2个品种鸡胸肌间共有134种DAMs,与AA肉鸡相比,48种DAMs在固始鸡胸肌中显著上调,86种DAMs显著下调,这些DAMs主要归属于氨基酸、脂肪酸和多肽.同时,基于胸肌转录组数据,筛选到1501个差异表达基因(differentially expressed genes,DEGs),与AA肉鸡相比,719个在固始鸡胸肌中上调,712个下调.代谢组和转录组联合分析显示,DEGs和DAMs在精氨酸和脯氨酸代谢、甘油磷脂代谢、甘氨酸及丝氨酸和苏氨酸代谢等通路显著富集.相关性分析显示,15个基因-代谢物分子对呈显著正相关,13个基因-代谢物分子对呈显著负相关,涉及L-丝氨酸、4-乙酰氨基-2-氨基丁酸、N2-琥珀酰-L-鸟氨酸、O-磷酸乙醇胺、胆碱等代谢物分子及GATM、SARDH、PTDSS2、ALDH7A1、AGPAT2和EHHADH等基因.[结论]与AA肉鸡相比,固始鸡中显著上调的差异代谢物(磷脂酰乙醇胺、L-丝氨酸、谷胱甘肽、肌肽、肌醇和苹果酸等)及其相关差异基因,可作为固始鸡胸肌肉质风味的潜在标志物.

[Objective]This study was conducted to explore the the characterization and potential regu-latory mechanisms of excellent meat quality traits in Gushi chickens.[Method]Liquid chromatography-mass spectrometry(LC-MS)was used to characterize the breast muscle metabolites of 120-day-old Gushi chickens using 45-day-old A A broilers as a control group,and the genes associated with the differential metabolite molecules were identified in conjunction with the breast muscle transcriptome da-ta.[Result]Metabolomic analysis showed that there were 134 differentially accumulated metabolite(DAMs)molecules between the two breeds of chicken breast muscle,and 48 DAMs were significantly up-regulated and 86 DAMs were significantly down-regulated in the breast muscle of Gushi chickens compared with AA broilers,which were mainly attributed to amino acids,fatty acids and peptides.Meanwhile,based on the breast muscle transcriptome data,1 501 differentially expressed genes(DEGs)were screened,and 719 were up-regulated and 712 were down-regulated in the breast muscle of Gushi chickens compared with AA broilers.Combined metabolomic and transcriptomic analyses re-vealed that DEGs and DAMs were significantly enriched in the pathways of arginine and proline metab-olism,glycerophospholipid metabolism,glycine and serine and threonine metabolism.Correlation anal-ysis showed that 15 gene-metabolite molecules pairs were significantly positively correlated and 13 gene-metabolite molecules pairs were significantly negatively correlated,involving L-serine,4-acetami-do-2-aminobutyric acid,N2-succinyl-L-ornithine,O-phosphoethanolamine,choline and other metabo-lite molecules and genes such as GATM,SARDH,PTDSS2,ALDH7A1,AGPAT2 and EHHADH,a-mong others.[Conclusion]Compared with AA broilers,significantly upregulated differential metabo-lites such as phosphatidylethanolamine,L-serine,glutathione,myostatin,myo-inositol,and malic acid and their related differential genes are potential markers of breast muscle flavor in Gushi chickens.
