Expression of LINC00641 in Colon Cancer and Its Effect on Biological Be-haviors of Colon Cancer Cells

Abstract
Objective:To analyze the correlation between the expression of LINC00641 in colon cancer tissue and the prognosis of colon cancer patients,and investigate the effects of LINC00641 on the biological behaviors of colon cancer cells and the epithelial-mesenchymal transition(EMT)process.Methods:Bioinformatics was used to analyze the correlation be-tween the expression of LINC00641 in human colon cancer tissue the prognosis of colon cancer patients.qRT-PCR was used to detect the expression of LINC00641 mRNA in SW480,HT29,HCT-116,SW620 and NCM460 cells.The expression of LINC00641 was knocked down using lentivirus vectors,and then MTT,Transwell,and flow cytometry were used to detect changes in cell proliferation,migration,invasion,apoptosis and cell cycle after interference.Western blot was used to detect the expression of EMT-related biomarkers,E-cadherin,N-cadherin,Twist1 and SOX2 proteins.Results:LINC00641 was highly expressed in colon cancer tissue(P<0.001);there was no significant difference in the overall survival between the LINC00641-low and LINC00641-high expression groups(P=0.377);and the disease-free survival in the LINC00641-high expression group was shorter than that in the LINC00641-low expression group(P=0.017).The expression of LINC00641 in colon cancer cell lines was significantly higher than that in normal colonic epithelial cell lines(P<0.05).After knocking down LINC00641 expression,the number of proliferated,migrated,and invaded cells significantly decreased;the cell cycle was arrested in the G0/G1 phase;and the rate of cell apoptosis significantly increased.Additionally,the relative expression level of E-cadherin protein increased,while the relative expression levels of N-cadherin,Twist1 and SOX2 proteins decreased(P<0.05).Conclusion:LINC00641 is highly expressed in colon cancer tissue and cell lines,and is associated with poor prognosis.High expression of LINC00641 affects the biological behaviors of colon cancer cells,potentially,which may en-hance the migration and invasion capabilities of colon cancer cells by promoting EMT.关键词
