Abstract

Objective:To investigate the effect of voltage-dependent anion channel 1(VDAC1)on breast cancer prolifer-ation and migration and its mechanism.Methods:The expression level and prognostic value of VDAC1 in breast cancer were detected using the TCGA database.The expression level of VDAC1 mRNA in breast cancer tissue and cell lines was assessed using reverse transcription quantitative polymerase chain reaction.The overexpression of VDAC1 in breast cancer cells was a-chieved through the use of an overexpressed plasmid,while the knockdown of VDAC1 expression was accomplished using small interfering RNA(si-RNA).The cell proliferation ability was assessed using CCK-8 and plate cloning assay,while the cell migration ability was evaluated through scratch assay and Transwell assay.The expressions of VDAC1,E-Cadherin,N-Cadherin,Vimentin,p-PI3K/PI3K,p-AKT/AKT and p-mTOR/mTOR proteins in cells were detected through Western blot.VDAC1-related genes were obtained from the TCGA database,and KEGG enrichment analysis was performed on these genes.Results:VDAC1 was highly expressed in breast cancer tissue and cell lines,and KEGG enrichment analysis showed that the PI3K-AKT-mTOR signaling pathway was significantly enriched.Overexpression of VDAC1 promoted the proliferation and mi-gration ability of SK-BR-3 cells,knockdown of VDAC1 inhibited the proliferation and migration ability of MDA-MB-231 cells.Western blot results showed that after overexpression of VDAC1,the expression of E-Cadherin in SK-BR-3 cells decreased,while the expressions of N-Cadherin,Vimentin,p-PI3K,p-AKT and p-mTOR increased;after the knockdown of VDAC1,the expression of E-Cadherin in MDA-MB-231 cells increased,while the expressions of N-Cadherin,Vimentin,p-PI3K,p-AKT and p-mTOR decreased.Conclusion:VDAC1 regulates the proliferation and migration of breast cancer through the PI3K-AKT-mTOR signaling pathway.
