Effects of aerobic exercise and resveratrol on the inflammation and apoptosis in kidney tissue of diabetes rats

探讨有氧运动和白藜芦醇对2型糖尿病大鼠肾组织炎症和凋亡的影响及其作用机制.选取SD大鼠65只,采用高糖高脂喂养结合化学诱导法建立糖尿病大鼠模型,将大鼠随机分为模型组、运动组、药物组和运动药物组,设立对照组.运动组大鼠完成连续8周的跑台运动,药物组大鼠采用白藜芦醇药液灌胃,每天给药1次,连续8周.8周结束,测血糖、24 h 尿白蛋白(24 hours urine protein,24 h-UP)定量、血肌酐(serum creatinine,Scr)和血尿素氮(blood urea nitrogen,BUN)等指标变化;TUNEL法检测肾组织细胞凋亡数量;ELISA检测与炎症反应有关白细胞介素-6(in-terleukin-6,IL-6)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)的表达;免疫组化法检测凋亡因子B淋巴细胞瘤-2(B cell lymphoma-2,Bcl-2)、Bcl-2相关X蛋白质(Bcl-2-associated X protein,Bax)、半胱天冬蛋白酶-3(cysteinyl aspartate specific proteinase 3,Caspase-3)的表达水平;蛋白免疫印迹(Western blot)法检测信号通路蛋白NF-κBp65的表达水平.结果显示,与对照组比较,模型组的Scr、BUN、血糖、24 h尿蛋白、炎症相关因子IL-6、IL-1β、TNF-α水平升高(P<0.05);肾脏细胞凋亡明显增多,肾脏Bax、Caspase-3、信号通路蛋白NF-κBp65表达显著上升,Bcl-2表达明显下降(P<0.05).分别施与有氧运动与白藜芦醇干预,运动组、药物组和运动药物组的Scr、BUN、血糖、24 h尿蛋白、IL-6、IL-1β、TNF-α水平均有不同程度的下降(P<0.05);肾脏细胞凋亡明显减少,Bax、Caspase-3、NF-KBp65表达显著下降(P<0.05),Bcl-2表达显著升高(P<0.05),且运动药物组的变化更明显.研究结果提示有氧运动、白藜芦醇单独或联合干预对糖尿病肾病大鼠肾脏有一定的保护作用,其机制可能与抑制NF-κBp65的活化,降低炎症因子水平,抑制肾脏细胞凋亡有关.有氧运动联合白藜芦醇干预减轻肾脏病理损伤的效果优于单一的有氧运动或白藜芦醇干预.

The aim of this study was to investigate the effects of aerobic exercise and resveratrol on renal inflammation and ap-optosis in type 2 diabetic rats and its mechanism.Sixty-five SD rats were selected to establish diabetic rat model by high glu-cose and high fat feeding combined with chemical induction.The rats were randomly divided into diabetic group,exercise group,drug group and exercise drug group,and normal control group.Exercise groups completed eight consecutive weeks of treadmill running.Drug groups were given resveratrol liquid once a day for eight weeks.At the end of eight weeks,blood glu-cose,24 h urinary albumin(24 h-UP),serum creatinine(Scr)and blood urea nitrogen(BUN)were measured.The number of apoptosis in renal tissue was detected by TUNEL method.The expressions of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)were detected by ELISA.The expressions of apoptosis factors B cell lymphoma-2(Bcl-2),Bcl-2-associated X protein(Bax)and cysteinyl aspartate specific proteinase 3(Caspase-3)were detected by immunohis-tochemistry.The expression of NF-KBp65 in renal was detected by Western blot.Compared with normal control group,the lev-els of Scr,BUN,blood glucose,24 h urinary protein,inflammation-related factors IL-6,IL-1β and TNF-α in model group were increased(P<0.05).Apoptosis of renal cells increased significantly,the expressions of Bax,Caspase-3 and NF-KBp65 in-creased significantly,and the expression of Bcl-2 decreased significantly(P<0.05).The levels of Scr,BUN,blood glucose,24 h urinary protein,IL-6,IL-1β and TNF-α in exercise group,drug group and exercise drug group were decreased to different degrees(P<0.05).The apoptosis of renal cells were decreased,the expressions of Bax,Caspase-3 and NF-KBp65 were sig-nificantly decreased(P<0.05),and the expression of Bcl-2 was significantly increased(P<0.05),and the changes were more obvious in the exercise drug group.The results indicated that aerobic exercise and resveratrol alone or combined inter-vention have a certain protective effect on diabetic nephropathy rats,and the mechanism may be related to inhibiting the acti-vation of NF-KBp65,reducing the level of inflammatory factors and inhibiting the apoptosis of kidney cells.Aerobic exercise combined with resveratrol intervention is better than single aerobic exercise or resveratrol intervention.
