国产便携式一体化人工心肺装置对急性呼吸窘迫综合征模型的作用机制研究
Study on the mechanism of domestic portable integrated artificial heart-lung device on acute respiratory distress syndrome model

Abstract
Objective The purpose of this study was to investigate the mechanism of new domestic portable integrated artificial heart-lung device in the treatment of acute respiratory distress syndrome(ARDS),alleviating systemic inflammatory response and improving endoplasmic reticulum stress injury.Methods Normal sheep were selected and set up as control group,ARDS model group and treatment group(n=10).The control group and the ARDS model group were given ulinastatin by intravenous bolus injection,and the treatment group was treated with a portable integrated artificial heart-lung device.The systemic inflammatory response,endoplasmic reticulum stress and other indicators in each group,as well as related indicators of lung tissue were abserved.Results The total cell number and polymorphonuclear white blood cell number in the alveolar lavage fluid of the treatment group were significantly lower than the ARDS model group,but significantly higher than that in the control group(P<0.05).The levels of inflammatory factors C-reactive protein,interleukin-1β,and tumor necrosis factor-α in the alveolar lavage fluid of the treatment group were significantly lower than those of the ARDS model group,but significantly higher than those of the control group(P<0.05),and the level of IL-10 was similar in contrast.The wet weight/dry weight value,apoptosis index,glucose regulatory protein 78,CCAAT/enhancer binding protein homologous protein,and caspase 12 index levels were higher in the ARDS model group,followed by the treatment group.The control group was the lowest.The nucleus in the alveolar epithelial cells of the control group were mainly light blue.In addition,the nucleus of the ARDS model group showed obvious pyknosis,and the cytoplasm was shrunk,and the nucleus of the treatment group was obviously relieved.Conclusion The new domestic portable artificial heart-lung can significantly improve the pulmonary stress response of the acute respiratory distress model,which is mainly related to the improvement of endoplasmic reticulum stress and the apoptosis of alveolar epithelial cells.关键词
