牛磺酸抑制心肌纤维化作用
Inhibitory Effect of Taurine on Myocardial Fibrosis

Abstract
Objective To investigate the effect of Taurine(Tau)on isoproterenol(Iso)induced myocardial fibrosis(MF)in rats and elucidate its possible mechanism.Method Fifty Wistar rats,weighing 200~250 g and half male and half female,were randomly divided into control group,model group(Iso group),low Tau(60 mg/(kg·d)),medium Tau(120 mg/(kg·d))and high Tau(240 mg/(kg·d))dose groups,with 10 rats in each group.The Iso rat MF model was established,the rats were injected with Iso 5 mg/(kg·d)in the back for 10 consecutive days.The control group was injected with normal saline of equal volume.Tau dose groups were intraperitoneally injected with Tau once a day from the 2nd day of modeling for 14 consecutive days.The heart mass index of each group was calculated.Myocardial histopathological changes were observed by HE staining.The collagen percentage of myocardial tissue was calculated by Masson staining.The mRNA expression of typeⅠ,type Ⅲ collagen and connective tissue growth factor(CTGF)in myocardial tissue was detected by qRT-PCR.Results The heart mass index in Iso group was significantly higher than that in control group(P<0.05).Compared with the model group,the heart mass index of rats in low,medium and high dose Tau groups was significantly decreased(P<0.05),which was concentration-dependent.HE staining showed that there were typical MF pathological changes in myocardial tissue of Iso group.The degree of the above pathological changes was significantly improved in the low,medium and high dose Tau group,and had a positive correlation with the concentration.Compared with control group,the percentage of myocardial collagen in Iso group was significantly higher than that in control group(P<0.05).Compared with Iso group,low,medium and high doses of Tau groups could significantly reduce the percentage of myocardial collagen in rats(P<0.05).The results of qRT-PCR showed that the expression levels of typeⅠ,type Ⅲ collagen and CTGF mRNA in myocardial tissue of Iso group were significantly higher than those of control group(P<0.01).Compared with the Iso group,low,medium and high doses of Tau groups could significantly reduce the expression levels of typeⅠand type Ⅲ collagen and CTGF mRNA(P<0.05).Conclusion Tau can significantly reduce the cardiac mass index of rats,improve the degree of cardiac tissue lesion,reduce the expression levels of typeⅠandⅢcollagen and CTGF mRNA in myocardial tissue,and inhibit MF in rats.关键词
