miRNA-200b在糖尿病小鼠海马体中的异常表达及靶基因预测

Abstract
Objective To analyze the expression of miRNA-200b in hippocampus of diabetes mice and predict the target gene of miRNA-200b.Methods Type 1 and type 2 diabetes mouse models were induced by streptozotocin or combined with high-sugar and high-fat diet,respectively.After modeling,the mice were killed and the hippocampus tissues were isolated.The expression of miRNA-200b was detected by fluorescence quantitative PCR.miRNA-200b target genes were predicted by miRWalk,miRDB and miRPathDB gene data-bases.Then,GO analysis and KEGG analysis were performed.Results The diabetes mouse model was successfully established,and the expression levels of miRNA-200b in the hippocampus of diabetic mice were decreased compared with that of normal mice(P<0.05).Bio-informatics prediction showed that three target genes of miRNA-200b were screened,namely GAB1,SLC5A3 and GICCL1.The functions of target genes were concentrated in the signaling pathways of glucose metabolism and neuronutrition.Conclusion The results of this study suggest that miRNA-200b is significantly decreased in the hippocampus of diabetic mice,which is expected to be a new target for diabetes treatment.关键词
