目的 观察小儿复方鸡内金咀嚼片对KM小鼠全身过敏反应、分离自SD大鼠腹腔液肥大细胞(RPMC)和Wistar大鼠嗜碱性白血病细胞株RBL-2H3类过敏反应的诱发结果.方法 ①取48只KM小鼠,随机均分为甲、乙、丙、丁组:甲组小鼠用小儿复方鸡内金咀嚼片混悬液灌胃,0.25 g生药/kg,隔天1次,连用3次,分别间隔9、16天时再次灌胃;乙组小鼠用小儿复方鸡内金咀嚼片混悬液灌胃,0.025 g生药/kg,隔天1次,连用3次,分别间隔9、16天时再次灌胃;丙组小鼠用1 mg卵清蛋白静脉注射,隔天1次,连用3次,分别间隔9、16天时再次静脉注射;丁组小鼠用0.1 mL/10 g生理盐水静脉注射,隔天1次,连用3次,分别间隔9、16天时再次静脉注射,观察各组小鼠主动全身过敏反应情况.②另取10只SD大鼠,分离大鼠腹腔原代RPMC细胞,分为5组:空白对照、C48/80组、高中低剂量组,高中低剂量组细胞分别加入32、16、8 mg/mL的小儿复方鸡内金咀嚼片混悬液,C48/80组及空白对照组分别加入0.04 mg/mL的组胺释放剂和肥大细胞脱颗粒剂C48/80、缓冲液,测算各组细胞组胺、β-氨基己糖苷酶释放率及促进率.③取对数生长期RBL-2H3细胞,分为空白对照、C48/80组、高中低剂量组,高中低剂量组细胞分别加入32、16、8mg/mL的小儿复方鸡内金咀嚼片混悬液,C48/80组及空白对照组分别加入0.04 mg/mL的C48/80、缓冲液,测算各组细胞组胺、β-氨基己糖苷酶释放率及促进率.结果 第14、21天时甲乙丁组小鼠均未出现过敏反应,过敏反应阴性;丙组小鼠出现呼吸困难、死亡,过敏反应强阳性.RPMC细胞C48/80组组胺释放率72.6%(致敏阳性),高中低剂量组的组胺释放促进率均低于64.23%(不具有致敏作用);C48/80组β-氨基己糖苷酶释放促进率为110.53%,(致敏阳性),高中低剂量组的β-氨基己糖苷酶释放促进率均低于80.31%(不具有致敏作用).RBL-2H3细胞C48/80组的组胺释放率促进率81.6%,高中低剂量组的组胺释放促进率均低于64.23%;C48/80组β-氨基己糖苷酶释放促进率为120.93%(致敏阳性),高中低剂量组β-氨基己糖苷酶释放促进率均低于80.31%(不具有致敏作用).结论 小儿复方鸡内金咀嚼片不诱发KM小鼠全身过敏反应,同时也不导致分离自SD大鼠腹腔原代肥大细胞及Wistar大鼠嗜碱性白血病细胞株RBL-2H3细胞的类过敏反应.

Objective To observe the induction results of Child Compound Endothelium Comeum(CCEC)on sys-temic allergic reactions in KM mice and anaphylactoid reaction in rat peritoneal mast cells(RPMC)and basophilic leuke-mia cell line RBL-2H3 in Wistar rats.Methods(1)Forty-eight KM mice were randomly divided into groups A,B,C and D.Mice in the group A were given 0.25g crude drug/kg CCEC suspension,once every other day for three consecutive times,and they were gavaged again at intervals of 9 and 16 days,respectively;mice in the group B were given 0.025 g crude drug/kg CCEC suspension,once every other day for three consecutive times,and they were gavaged again at inter-vals of 9 and 16 days,respectively;mice in the group C were given 1 mg ovalbumin(OVA)intravenously,once every oth-er day for three consecutive times,and they were gavaged again at intervals of 9 and 16 days,respectively;mice in the group D were given intravenous injection of 0.1 mL/10 g normal saline,once every other day for three consecutive times,and they were gavaged again at intervals of 9 and 16 days,respectively;the active systemic allergic reaction of rats in each group was observed.(2)Primary RPMC were isolated from the abdominal cavity of another 10 SD rats and were divided in-to five groups:the blank control group,C48/80 group,high-,medium-and low-dose groups;cells in the high-,medi-um-,and low-dose groups were added with 32,16 and 8 mg/mL of CCEC suspension,respectively.The cells of the C48/80 group and blank control group were added with 0.04 mg/mL of histamine release agent and mast cell degranulation agent C48/80 and buffer solution,respectively;the release rate and promotion rate of histamine and β-amino-hexosidase were measured in each group.(3)RBL-2H3 cells in the logarithmic growth phase were divided into the blank control group,C48/80 group and high-,medium-and low-dose groups.The cells in high-,medium-and low-dose groups were added with 32,16 and 8mg/mL of CCEC suspension,respectively,and the cells in C48/80 group and blank control group were added with 0.04 mg/mL of C48/80 and buffer solution,respectively.The release rate and promotion rate of hista-mine and β-amino-hexosidase were measured in each group.Results On the 14th and 21st days,no anaphylaxis oc-curred in the groups A,B,and D,and the anaphylaxis was negative.The mice in the group C had dyspnea,death and strong positive allergic reaction.The release rate of histamine in the C48/80 group was 72.6%and the release rates and promotion rates of histamine in the high-,medium-and low-dose groups were lower than 64.23%.The release rate and promotion rate of β-aminoglycosidase were 110.53%in the C48/80 group(positive sensitization),and the release rates and promotion rates of β-aminoglycosidase were lower than 80.31%in the high-,medium-and low-dose groups(no sensi-tizing effect).The release rate and promotion rate of histamine in the RBL-2H3 cells of the C48/80 group were 81.6%,and the release rates and promotion rates of the high-,medium-and low-dose groups were lower than 64.23%.The re-lease rate and promotion rate was 120.93%in the C48/80 group(positive sensitization),and the release rates and promo-tion rates of β-aminoglycosidase were lower than 80.31%in the high-,medium-and low-dose groups(no sensitizing ef-fect).Conclusions CCEC neither induced systemic allergic reactions in KM mice nor caused anaphylactoid reactions in primary RPMC and RBL-2H3 cells of basophilic leukemia cell line from Wistar rats.
