摘要

Abstract

Objective:To evaluate the causal relationship between high blood pressure,atrial fibrillation,and Alzheimer's disease using a dual sample Mendelian randomization of genetic variation,in order to avoid the limitations of observational studies.Methods:The data comes from the IEU Large high blood pressure(HBP)Dataset,atrial fibrillation(AF)Dataset,and Alzheimer's disease(AD).The causal effects were estimated using inverse variance weighting(IVW),mendelian randomization(MR)Egger,weighted median,Simple mode,and Weighted mode.Sensitivity analysis was conducted using Cochran's Q test and MR Egger intercept test.Re-sults:There was no significant association between genetically predicted HBP and the risk of AD(OR=0.89,95%CI:0.53~1.49,P=0.58),MR Egger did not find evidence of genetic pleiotropy(intercept=0.007,P=0.39).There was no association between AF and the risk of AD(OR=1.04,95%CI:0.97~1.10,P=0.75),MR Egger did not reveal evidence of genetic pleiotropy(intercept=0.002,P=0.63).Conclusion:Mendelian randomization analysis did not find a causal relationship between HBP,AF and AD.
