Effect of circ_0013958/miR-545 axis on the proliferation,apoptosis and migration of endometrial cancer cell RL-95-2

Abstract
Objective To explore the effects of circ_0013958 on the proliferation,apoptosis and migration of endometrial cancer cell RL-95-2 and its possible mechanism.Methods Endometrial cancer tissues and paracancerous tissues of 51 cases were collected,and the expression levels of circ_0013958 and miR-545 in the tissues were detected by quantitative real time polymerase chain reaction(qRT-PCR).RL-95-2 cells were cultured in vitro and transfected with sh-circ_0013958,miR-545 mimics,si-circ_0013958 and miR-545 inhibitor respectively,and then cell proliferation,apoptosis,and migration were detected by CCK-8,flow cytometry,and Transwell assays,respectively.The levels of apoptosis-related proteins were detected by Western blot.The targeting relationship between circ_0013958 and miR-545 was verified by the dual luciferase reporter assay.Results Compared with paracancerous tissues,the expression level of circ_0013958 in endometrial cancer tissues was higher(P<0.05),and the expression level of miR-545 was lower(P<0.05).After knocking down circ_0013958 or up-regulating miR-545,the proliferation and migration ability of RL-95-2 cells was inhibited,while cell apoptosis was increased(P<0.05).circ_0013958 targeted negative regulation of miR-545.Down-regulation of miR-545 reversed the anti-proliferation,anti-migration and pro-apoptosis effects mediated by knocking down circ_0013958.Conclusion circ_0013958 may promote the proliferation and migration of endometrial cancer RL-95-2 cells and hinder cell apoptosis by targeting down-regulation of miR-545.关键词
