Abstract

Leaf and stem segments of Rhodiola sachalinensis A.Bor were used as experimental materials to study callus induction and growth of Rhodiola sachalinensis under different mass concentrations of hormones,and non-targeted metabolomics analysis was performed based on GC-MS technology to comprehensively evaluate the differential metabolites of leaves and roots.The results showed that the optimal treatment conditions for leaf induction were MS+3.0 mg·L-1 NAA+7.0 mg·L-1 6-BA,and the healing rate reached 87.67%.The optimal treatment conditions for stem were MS+1.0 mg·L-1 NAA+5.0 mg·L-1 6-BA,and the healing rate reached 86.33%.The best hormone combinations for callus growth of leaves and stems were MS+0.5 mg·L-1 NAA+7.0 mg·L-1 6-BA.Stem segments showed stable and high levels during callus induction and callus growth.A total of 187 metabolites were isolated from the roots and leaves of Rhodiola sachalinensis,and 11 different metabolites were screened out.The metabolites with higher content in leaves are acids,while those with higher content in roots are sugars and alcohols.The experimental results are expected to provide theoretical experience for the development and utilization of local Rhodiola sachalinensis,and provide scientific guidance for further expanding the natural quantity and promoting the development of local Rhodiola sachalinensis industry.
