糖尿病肾病(diabetic kidney disease,DKD)的发病与TLR4 信号通路密切相关.中医药防治DKD具有多靶点、多效应、多成分等优势,取得了较好的临床疗效,主要通过靶向TLR4 信号通路从而抑制肾脏炎症反应、减轻氧化应激、改善肾脏纤维化、减少细胞凋亡等实现的.TLR4 信号通路已成为中医药防治DKD的重要靶点.但目前中医药防治DKD相关研究仍以基础实验为主,缺少高质量大样本临床试验验证;多为单一信号通路研究,中医药防治DKD的分子机制尚未阐明,需进一步挖掘TLR4 等多通路间的联系及交叉研究,阐明多靶点、多通路的作用机制,亦可利用网络药理学、多组学联合分析等技术,深入挖掘中药有效活性成分及其作用机制,以期为中医药防治DKD提供新的研究思路.

The TLR4 signaling pathway is closely related to the pathogenesis of diabetic kidney disease(DKD).TCM has achieved good clinical efficacy in the prevention and treatment of DKD with some advantages such as multiple targets,effects,and components.TCM can inhibit the kidney inflammation,reducing oxidative stress,improve renal fibrosis and decrease cell apoptosis by regulating TLR4 sig-naling pathway,which has become an important target for the prevention and treatment of DKD with TCM.However,at present,the re-search related to the prevention and treatment of DKD by traditional Chinese medicine is still mainly based on basic experiments,and there is a lack of high-quality large-sample clinical trial verification.The existing research are mostly focused on single signaling pathway,and the molecular mechanism has not been clarified.From this prospect,it is necessary to further explore the connection and intersection of multiple pathways such as TLR4 to clarify mechanism of the multi-target and multi-pathway,as well as the active com-ponents of TCM and their mechanism by using network pharmacology and multi-omics joint analysis technology,so as to provide new i-deas for the prevention and treatment of DKD with TCM.
