为了提高乳酸肠球菌在真空冷冻干燥过程中的存活率以及后期菌粉保存的效果,采用单因素试验和Box-Behnken响应面试验相结合的方法,筛选和优化乳酸肠球菌真空冷冻干燥过程中的最优保护剂配方.结果表明:脱脂奶粉、海藻糖和甘油3种保护剂效果较好,能够显著提高乳酸肠球菌真空冷冻干燥后的存活率.将上述3种保护剂作为自变量,以冻干后菌体的存活率作为响应值进行响应面试验,得到乳酸肠球菌优化保护剂配方:脱脂奶粉20.82%、海藻糖13%、甘油3%,通过进一步验证和保存试验研究,采用优化后的保护剂配方,菌体冻干后的存活率均在80.00%左右,冻干菌粉在4 ℃条件下可以保存24个月.

In order to improve the survival rate of Enterococcus lactate and the preservation effect of bacterial powder in later period during vacuum freeze-drying,the single factor experiment and Box-Be-hnken design was used to screen and optimize the protective agents formula for vacuum freeze-drying of Enterococcus lactate.The results showed that the skim milk,trehalose and glycerin have the best effect,which can significantly improve the survival of Enterococcus lactate after vacuum freeze-drying.In response surface experiment of Box-Behnken,the above three protective agents were taken as independent varia-bl es,and the survival rate of cells after lyophilization was the response value.The optimized protective a-gent formula of Enterococcus lactate was obtained:skim milk 20.82%,trehalose 13%,glycerin 3%.Fur-ther verification and stability research showed that the survival rate of lyophilized thallus reached about 80.00%with the optimized protective agent formula,the lyophilized Enterococcus lactate powder can be preserved for 24 months at 4 ℃.
