鸭绿江花羔红点鲑的DNA条形码及鉴定分析
DNA Barcode and Identification Analysis of Salvelinus malma in Yalu River
摘要
Abstract
The purpose of this study was to identify the species of Dolly Varden Trout in Yalu River by DNA barcoding technique.Through analyzing the gene sequence of cytochrome oxidase Ⅰ subunit(CO Ⅰ)of Yalu River Dolly Varden Trout,the species of Yalu River Dolly Varden Trout was identified by DNA barcoding.The results showed that the length of mitochondrial CO Ⅰ gene was 699 bp,and the contents of A,T,C and G bases were 22.75％,29.61％,28.76％and 18.88％,respectively,in which the content of G was lower,the content of A+T(52.36％)was higher than that of C+G(47.64％).There were no significant intraspecific differences and the average intraspecific genetic distance of the Dolly Varden Trout was less than 0.02.DNA gene sequence analysis showed that the highest similarity with the GenBank and Bold Systems was 99.56％-100.00％.A comprehensive analysis of intraspecific genetic distance and search results showed Dolly Varden Trout in the Yalu River was Salvelinus malma.关键词
