橡胶树白粉菌分生孢子DNA的微量提取方法建立OA北大核心CSTPCD
Establishment of a Microscale DNA Extraction Method for Rubber Tree Powdery Mildew Spores
由橡胶树白粉菌(Erysiphe quercicola)引发的橡胶树白粉病是我国天然橡胶产业中的一个重要病害,其对乳胶产量造成了严重威胁.随着预测预报技术的不断进步,结合qPCR检测和孢子捕捉技术有望成为高效预测白粉病流行的新方法.本研究针对橡胶树白粉菌分生孢子DNA的微量提取,通过比较不同预处理方式、不同材质的研磨珠以及不同类型的研磨装置,筛选出了最适合的孢子样品研磨体系.进一步比较了 CTAB法与几种商业提取试剂盒的提取效率,最终确定了一种可行的孢子DNA微量提取方法:将白粉菌分生孢子水溶液置于4℃超声处理10 min后加入2 mm氧化锆珠0.2 g、0.4~0.6 mm玻璃珠0.2 g以及0.2 mm氧化锆珠0.2 g,用组织研磨仪在4 ℃ 70 Hz条件下研磨40 s,停10s,20个循环,最后使用CTAB法提取DNA.通过qPCR定量,可以在101个/mL的孢子悬浮液当中检出橡胶树白粉菌DNA,且不受田间复杂情况的干扰,具有高灵敏度和强特异性,为橡胶树白粉病的预测预报提供了可靠的理论依据和有效的技术手段.
Powdery mildew caused by Erysiphe quercicola is a significant disease in China's natural rubber industry,posing a severe threat to latex production.With the continuous advancement of predictive and forecasting technologies,the combination of qPCR detection and spore capture techniques holds promise as an efficient method for predicting the prevalence of powdery mildew.In this study,we focused on the microscale extraction of DNA from rubber tree powdery mildew spores.We compared various pre-treatment methods,different types of grinding beads with varying materials,and different types of grinding machines to identify the most suitable spore sample grinding system.Furthermore,we compared the efficiency of the CTAB method with several commercial extraction kits.Ultimately,we established a fea-sible microscale DNA extraction method for spores:first,the powdery mildew spore water solution subjected to 4 ℃ ultrasonication for 10 min,subsequently,2 mm zirconia beads(0.2 g),glass beads(0.4-0.6 mm,0.2 g),and 0.2 mm zirconia beads(0.2 g)added,tissue grinder 70 Hz at 4 ℃,grind 40 s,stop 10 s,20 cycles,finally,DNA extracted using the CTAB method.This method effectively and stably extracts DNA from 101 spores/mL rubber tree powdery mildew spores,without being affected by complex field conditions,and demonstrates high sensitivity and specificity.It provides a reliable theoretical basis and an effective technical approach for the prediction and forecasting of rubber tree powdery mildew.
