Abstract
Objective To identify key genes associated with metastasis in colon cancer and their clinical significance.Methods Based on colon cancer datasets from TCGA and GEO(GSE41258),we used the WGCNA and Limma packages to ana-lyze gene sets related to colon cancer metastasis,and studied the impact of these gene sets on prognosis and clinical tumor staging,with statistically significant genes identified as key genes.Subsequently,using Metascape,we constructed protein interaction net-works for each key gene,and performed functional enrichment analysis on these networks with DAVID.Lastly,by utilizing the TIDE database,we analyzed the influence of key genes on immune infiltration by cytotoxic T cells and the immune response from an im-munological perspective.Results The study identified six key genes:CNN1,CXCL10,CXCL11,MGP,SPINK4,and PBK,which not only affected patient prognosis but also showed a linear relationship with the stages of colon cancer.Through KEGG and GO en-richment analysis,it was found that the key gene networks regulating the immune processes such as cytokine activity,chemotaxis of centriole cells,and chemotaxis of monocytes were implicated in liver metastasis of colon cancer,while in lung metastasis,the DA-VID RAS signaling pathway was indicated.These results revealed the mechanistic differences in different metastatic pathways of co-lon cancer.Among them,CNNl and MGP suppressed the immune response of cytotoxic T cells,while PBK promoted it.Conclusion The six genes,CNN1,CXCL10,CXCL11,MGP,SPINK4,and PBK,can serve as molecular biomarkers for the screening of colon cancer metastasis and play a role in the immunotherapy of colon cancer,presenting potential therapeutic targets for treatment.关键词
