针刺足三里对小鼠术后肠麻痹的作用机制OACSTPCD

目的:本研究旨在探索针刺足三里穴位治疗术后肠麻痹的作用及其信号传导机制.方法:使用野生型小鼠和Toll样受体4基因敲除小鼠各10只构建术后肠麻痹模型.比较Toll样受体4基因敲除小鼠、野生型小鼠术后胃肠运动功能、肠黏膜损伤情况.使用20只野生型小鼠构建术后肠麻痹模型,随机分为电针足三里穴位组和足三里穴位对照组,每组10只,电针足三里穴位组在造模后6、24、48、72 h连续足三里穴位电针刺激4次,足三里穴位对照组连接电针仪但不给予电刺激,检测两组实验动物胃肠功能运动情况、小肠组织炎症因子水平和Toll样受体相关通路关键分子TLR4、TRAF6及JNK表达.结果:与野生型小鼠相比,Toll样受体4基因敲除小鼠的胃肠运动功能改善,小肠组织炎症因子表达下调,肠黏膜损伤较轻.给予电针足三里穴位刺激可改善胃肠运动功能,减轻小肠组织炎症因子表达水平,并下调Toll样受体4相关因子TLR4、TRAF6和JNK表达水平.结论:电针足三里穴位可减轻术后肠麻痹,Toll样受体4信号通路在其中可能发挥关键调节作用.

Objective:Study aims to explore effects of acupuncture at Zusanli acupoint on postoperative ileus and its information transmission mechanism.Methods:Ten wild-type mice and ten Toll like receptor 4 gene knockout mice were used to construct postoperative ileus models.The postoperative gastrointestinal motility and intestinal mu-cosal damage were compared.Twenty wild-type mice were used to construct postoperative ileus model,which were randomly divided into electroacupuncture Zusanli acupoint group and control group,with ten mice in each group.The electroacupuncture Zusanli acupoint group were continuously stimulated with electroacupuncture at Zusanli acupoint 4 times at 6 h,24 h,48 h,and 72 h after modeling.The control group were connected to electroacupuncture instru-ment but not given electrical stimulation.The gastrointestinal motility function,levels of inflammatory factors in small intestine tissue,and key molecule Toll like receptor 4 related to Toll like receptor pathways in both groups were detected TRAF6 and JNK expression.Results:Compared with wild-type mice,Toll like receptor 4 gene knockout mice showed improved gastrointestinal motility,down-regulated expression of inflammatory factors in small intestine tissue,and milder intestinal mucosal damage.Stimulating Zusanli acupoint with electroacupuncture could improve gastrointestinal motility,reduce expression levels of inflammatory factors in small intestine tissue,and down-regulate the expression levels of Toll like receptor 4 related factorsTLR4,TRAF6 and JNK.Conclusion:Electroacupuncture at Zusanli acupoint could alleviate postoperative intestinal paralysis,and the Toll like receptor 4 signaling pathway might play key regulatory role in the process.
