基于JAK2-STAT3信号通路探讨热毒宁注射液治疗大鼠急性肺损伤的作用机制
Mechanism of Reduning injection in improving acute lung injury in rats based on the JAK2-STAT3 signaling pathway

Abstract
Objective:To investigate the mechanism of Reduning injection in the intervention of acute lung injury(ALI)rats by regulating JAK2-STAT3 signaling pathway.Methods:30 Wistar rats were randomly divided into model control group,blank control group,dexamethasone group,Reduning high-dose group and low-dose group separately.The administration method is intraperitoneal injection once a day for 5 consecutive days.After the last administra-tion,the ALI rat model was established by lipopolysaccharide infusion into the tracheal wall of posterior pharynx.24 h after making the model,the arterial oxygen pressure was compared among all groups,the pathological changes of lung tissue were observed by HE staining,and the expression levels of TNF-α,IL-1βand IL-6 in lung tissue were detected by ELISA.The protein expression levels of JAK2,p-JAK2,STAT3 and p-STAT3 in lung tissues of each group were determined by Western blot.Results:Compared with blank control group,PaO2 level of model control group was significantly decreased,pathological score was increased,PaCO2,IL-1β,IL-6,TNF-αlevel,JAK2,p-JAK2,STAT3 and p-STAT3 protein expression were increased(all P<0.05).Compared with model control group,the PaO2 level in dexamethasone group and Reduning high-dose group were increased,pathological score,PaCO2,IL-1β,IL-6,TNF-αlevel,JAK2,p-JAK2,STAT3,p-STAT3 protein expression were decreased(all P<0.05).Conclusion:Reduning injection has a protective effect on ALI rats,and its mechanism may be related to regulating the JAK2-STAT3 signaling pathway and reducing inflammatory response.关键词
