Genetic diversity analysis of 27 compositae germplasms based on ISSR molecular markers

采用ISSR分子标记技术对27份菊科种质进行亲缘关系和聚类分析,为种质资源的有效保存及分子鉴定奠定基础.利用NTSYS-pc 2.1软件对电泳结果进行多态性分析,采用非加权平均距离法(UPGMA)构建遗传树.结果表明:10条引物扩增出255条清晰的多态性条带,占总条带数的94.80%.UPGMA聚类分析显示,27份菊科种质间的遗传相似系数为0.36-1.00,在相似系数0.40处,27份菊科种质被划分为3个类群.目前搜集的27份菊科植物种质遗传多样性较为丰富,筛选的10个ISSR引物可进一步用于后续种质资源的分子鉴定.

In order to preserve and identify compositae germplasm resources,ISSR markers were used to analyze the genetic diversity and genetic relationship of 27 compositae germplasms.The polymorphisms were analyzed by NTSYS-pc 2.1,the genetic tree was constructed by UPGMA.The results showed that 10 primers were screened and 255 clear polymorphic bands were amplified,accounting for 94.80%of the total bands.The UPGMA cluster analysis showed that the genetic similarity coefficients of 27 compositae germplasms ranged from 0.36 to 1.00,and the 27 compositae germplasms could be divided into three categories at 0.40 similarity coefficient.The 27 compositae germplasms have rich genetic diversity and the 10 ISSR primers can be used for molecular identification of germplasm resources.
