两种昆虫生长调节剂类杀虫剂对甜菜夜蛾幼虫储存蛋白含量的影响
Effects of two insect growth regulator insecticides on storage protein content of Spodoptera exigua larvae

Abstract
The storage protein of the 5th instar larvae of Spodoptera exigua was purified by anion exchange and gel filtration.SDS-PAGE and ELISA were used to determine the dynamic changes of storage protein in the 5th instar,and to study the effects of two insect growth regulator insecticides,methoxycarbazide and chlorfluazuron,on the storage protein content at sublethal doses.The results showed that the molecular weight of the storage protein subunit in larvae of S.exigua was 85.1 ku.In the hemolymph of 5th instar larvae,the storage protein concentration gradually increased with day-age and reached its maximum value of 30.76 mg/mL on the 3rd day of 5th instar.Low doses of methoxycarbazide could increase the storage protein concentration in the hemolymph of larvae.After 48 h and 72 h of treatment,the storage protein concentration in the LC10 treatment group was 19.27 mg/mL and 45.21 mg/mL,significantly higher than that in the control group.High doses of methoxycarbazide had a significant inhibitory effect on the storage protein in S.exigua.After 72 h,the storage protein concentration in the hemolymph of the LC40 treatment group was only 48.47％of that of the control group.The storage protein concentration in the hemolymph of the LC10and LC40 treatment groups was lower than that of the control group at all treatment times,especially after 48 h and 72 h of treatment,the storage protein concentration in the hemolymph significantly decreased.The higher the concentration of chlorfluazuron,the more significant the effect.After 72 h of treatment,the storage protein concentration in the hemolymph of the LC40 treatment group was only 26.77％of that of the control group.The research results could provide a theoretical basis for further revealing the toxic effects of two insecticides on S.exigua larvae.关键词
