目的:探索鼻敏宁颗粒对寒证变应性鼻炎大鼠鼻黏膜组织中黏蛋白5B(MUC5B)、黏蛋白5AC(MUC5AC)、Toll样受体2(TLR2)蛋白表达情况.方法:将60只大鼠随机分成空白对照组、模型组、阳性对照组、鼻敏宁低、中、高剂量组.造模成功后,鼻敏宁颗粒高剂量组大鼠予鼻敏宁颗粒2 g/kg,鼻敏宁颗粒中剂量组1 g/kg,鼻敏宁颗粒低剂量组0.5 g/kg,阳性对照组予辛芩颗粒5 g/kg,2 mL/次,空白对照组与模型组给予同等容积的0.9%氯化钠注射液灌胃,均为每天1次,连续7 d.应用免疫组织化学定位检测鼻黏膜上皮细胞TLR2蛋白表达情况,应用蛋白质印迹法(WB)检测鼻黏膜上皮组织MUC5B及MUC5AC蛋白表达量.结果:鼻敏宁低中高剂量组中阳性染色明显减弱,模型组与其他组比较,差异有统计学意义(P<0.05);与模型组比较,中剂量组与高剂量组MUC5B及MUC5AC含量显著降低,有明显的差异有统计学意义(P<0.05).结论:鼻敏宁颗粒可通过降低黏蛋白5B及黏蛋白5AC的表达,TLR2蛋白的表达,减轻大鼠鼻黏膜免疫损伤,达到治疗变应性鼻炎的作用.

Objective:To explore the expression of mucin 5B(MUC5B),mucin 5AC(MUC5AC),and Toll-like receptor 2(TLR2)in the nasal mucosa of the rats with allergic rhinitis(AR)due to cold syndrome and treated with Biminning Granules.Methods:Sixty rats were randomized into 6 groups:blank control,model,positive control(Xinqin Granules,5 g/kg,2 mL per time),and low-,medium-,and high-dose(0.5,1,and 2 g/kg,respectively)Biminning Granules.The successfully modeled rats were administrated with corresponding drugs or 0.9%sodium chloride(blank control and model groups)by gavage once a day for 7 days.The immu-nohistochemical assay was employed to localize the expression of TLR2 in the nasal mucosa.Western blotting was employed to de-termine the expression levels of MUC5B and MUC5AC in the nasal mucosa.Results:Positive staining was significantly attenuated in the Biminning groups,and the model group showed significant differences compared with other groups(P<0.05).Compared with the model group,medium and high-dose Biminning groups showcased down-regulated expression of MUC5B and MUC5AC(P<0.05).Conclusion:Biminning Granules can reduce the immune damage to the nasal mucosa by down-regulating the expres-sion of MUC5B,MUC5AC,and TLR2,thereby treating AR in rats.
