抗菌肽CC34对小鼠肠炎的缓解作用
Alleviating effect of antimicrobial peptide CC34 on mouse enteritis

Abstract
To reveal the alleviating effect and mechanism of antimicrobial peptide CC34 on lipopolysaccharide(LPS)-induced intestinal inflammation in mice,the experiment selected 40 SPF mice,randomly divided into four groups,with ten mice in each group.Mice in the CC34 group and the LPS+CC34 group were intraperitoneally injected with 9 mg/kg CC34,while mice in the LPS group and Control group were injected with an equal volume of saline,for a duration of seven days.The mice in the LPS group and LPS+CC34 group were injected with 7.5 mg/kg LPS,the CC34 group and Control group injected with equal volume normal saline.The results showed that compared to the Control group,there were no significant changes in the liver,spleen,and heart indices of mice in the CC34 group(P>0.05),but the colon length extremely increased(P<0.01).Compared to the LPS group,the bacterial load in the liver and the content of inflammatory factors in the duodenum and ileum of mice in the LPS+CC34 group extremely decreased(P<0.01),and the gene expression levels of ZO-1,Claudin-1,and Occludin in the intestinal tissue extremely increased(P<0.01),and the villus height/crypt depth ratio(V/C)in the duodenum significantly increased(P<0.05).The gene expression level of myeloperoxidase(MPO)in the intestinal tissue extremely decreased(P<0.01).The gene expression and activity of superoxide dismutase(SOD)and catalase(CAT)in the intestinal tissue extremely increased(P<0.01),the content of malondialdehyde(MDA)extremely decreased(P<0.01),and the total antioxidant capacity(T-AOC)extremely enhanced(P<0.01).The gene expression levels of toll-like receptor-4(TLR-4)and myeloid differentiation factor 88(MyD88)in the intestinal tissue extremely down-regulated(P<0.01),and the phosphorylation level of p65 in the duodenum extremely decreased(P<0.01).The study indicates that CC34 alleviates intestinal inflammation in mice by down-regulating the TLR-4/NF-κB signaling pathway.关键词
