变构蛋白的调控机制及其分子设计的研究进展OACSTPCD
Advances in the mechanism of allosteric proteins and their molecular design
变构蛋白是一类具有特殊变构效应的蛋白质,其变构动态调节的多样性大大丰富了蛋白质的活性或特异性.探究蛋白变构效应及其调控机制,在拓展蛋白功能、药物研发和信号传递等方面具有重要的意义.本文从变构蛋白的功能与分类、调控机制、结构解析和变构位点的理性预测、人工变构蛋白的改造等方面进行总结,综合分析了开-关变构调控、变构协调调控和间接变构调控这 3 种主要的变构蛋白的调控机制、研究难点以及对应策略,从而为变构蛋白的结构理性设计提供相应的理论支持.
Allosteric proteins are a class of proteins with specific allosteric effects,and the exploration of their variousness allosteric mechanisms greatly enriches the activity and selectivity of the functional library of proteins.The investigation of the allosteric effects and their regulatory mechanisms is of great significance for the expansion of protein functions,drug discovery and signal transmit.We summarize recent the functions and classification of allosteric proteins,regulatory mechanisms,structural analysis,rational prediction of allosteric sites,and modification of artificial allosteric proteins,and comprehensively analyze the On-Off conformational regulation,cooperative allosteric regulation,and indirect allosteric regulation mechanisms of allosteric proteins,current research difficulties and possible corresponding strategies so as to provide theoretical support for the rational design of allosteric protein.
