TRIM59对人下咽癌细胞增殖、凋亡、侵袭和迁移的影响OACSTPCD
Effect of TRIM59 on proliferation,apoptosis,invasion and migration of human hypopharyngeal cancer cells
目的 探讨三结构域蛋白59(tripartite motif containing 59,TRIM59)对下咽癌细胞Fadu的生物学行为的影响及其机制.方法 通过肿瘤免疫估计资源数据库2.0(TIMER2)了解TRIM59在不同类型癌症中的表达情况,初步评估其在头颈部肿瘤中的表达差异.培养下咽癌细胞株Fadu,随机分为:实验组(si-TRIM59-1组,si-TRIM59-2组,si-TRIM59-3组)和对照组(NC组).通过实时定量PCR法检测转染效率,选取转染成功且基因下调效率最高的两组进行后续实验.CCK-8实验、集落形成实验检测Fadu细胞增殖能力;Transwell实验、细胞划痕实验检测Fadu细胞迁移及侵袭能力;Western blot实验检测下调TRIM59后,TRIM59、JAK2/pJAK2、STAT3/pSTAT3、Bcl-2、Bax的表达;流式细胞术检测Fadu细胞凋亡率.结果 与NC组比较,实验组TRIM59 mRNA相对表达量下降(P<0.001),其中,si-TRIM59-1组和si-TRIM59-3组下降更为明显(P<0.05),因此,选取si-TRIM59-1组和si-TRIM59-3组进行后续实验.与NC组比较,si-TRIM59-1组和si-TRIM59-3组细胞增殖、迁移及侵袭能力均下降(P<0.05).与 NC 组比较,si-TRIM59-1 组和 si-TRIM59-3 组 TRIM59、JAK2/p-JAK2、STAT3/p-STAT3 表达量降低(P<0.05),凋亡相关蛋白中抑制蛋白Bcl-2表达量下降,而诱导蛋白Bax表达量则上升(P<0.05).与NC组比较,si-TRIM59-1组和si-TRIM59-3组细胞凋亡率上升(P<0.05).结论 下调TRIM59后,人下咽癌细胞株Fadu的增殖、迁移及侵袭能力下降,细胞凋亡率上升,其机制可能与JAK2/STAT3通路有关.
Objective To investigate the effects of tripartite motif containing 59(TRIM59)on the biological behaviors of hypopharyngeal cancer cells Fadu and its mechanism.Methods The expression of TRIM59 in different types of cancer was investigated through the Tumor Immune Estimation Resource database 2.0(TIMER2),and its expression difference in head and neck tumors was initially eva-luated.Nasopharyngeal carcinoma Fadu cells were cultured and randomly divided into experimental group(si-TRIM59-1 group,si-TRIM59-2 group,si-TRIM59-3 group)and control group(NC group).Real-time quantitative PCR was used to detect the transfection efficiency,and the two groups with successful transfection and the highest efficiency of gene downregulation were selected for subse-quent experiments.The proliferation ability of Fadu cells was detected by CCK-8 assay and colony formation assay.The migration and invasion abilities of Fadu cells were detected by Transwell assay and cell scratch assay.After TRIM59 was downregulated,the expressions of related proteins(TRIM59,JAK2/p-JAK2,STAT3/p-STAT3,Bcl-2,Bax)were detected by Western blot assay.The apoptosis rate of Fadu cells was detected by flow cytometry.Results Compared with NC group,the relative mRNA expression of TRIM59 was decreased in si-TRIM59-1 group,si-TRIM59-2 group and si-TRIM59-3 group(P<0.001),especially si-TRIM59-1 group and si-TRIM59-3 group(P<0.05).So si-TRIM59-1 and si-TRIM59-3 were chosen for subsequent experiments.Compared with NC group,the proliferation,migration and invasion abilities of Fadu cells in si-TRIM59-1 group and si-TRIM59-3 group were decreased(P<0.05).Compared with NC group,the expression levels of TRIM59,JAK2/p-JAK2,STAT3/p-STAT3 were decreased in si-TRIM59-1 group and si-TRIM59-3 group(P<0.05),and the expression of inhibitory protein Bcl-2 was decreased while the expression of inducible protein Bax was increased(P<0.05).Compared with NC group,the apoptosis rate was increased in si-TRIM59-1 group and si-TRIM59-3 group(P<0.05).Conclusion TRIM59 downregulation can inhibit the proliferation,migration and invasion of human hypopharyngeal cancer cell line Fadu,and induce the apoptosis rate,which may be related to the JAK2/STAT3 pathway.
