Effects of polydatin on apoptosis of cervical cancer cells and transplanted tumor in nude mice by JAK1/STAT3 signaling pathway

Abstract
Objective:To investigate the effect of polydatin on apoptosis of cervical cancer cells and transplanted tumor mice by regulating Janus kinase 1(JAK1)/signaling and transcriptional activator 3(STAT3)signaling path-way.Methods:Human cervical cancer HeLa cell lines were selected and divided into control group(HeLa cells with-out any treatment)and polydatin low,medium and high dose groups(50,100 and 150 μmoL/L polydatin were add-ed).The culture was continued for 24,48 and 72 hours.Annexin V double staining was used to detect the apoptosis of HeLa cells.The proliferation ability of HeLa cells was detected by MTT,the invasion ability of HeLa cells was de-tected by Transwell chamber,the mRNA expression of JAK1 and STAT3 was detected by qRT-PCR,and the expres-sion of JAK1/STAT3 signaling pathway related proteins was detected by Western blot,and the cell morphology was ob-served.Results:Compared with the control group,the apoptosis rate of HeLa cells in polydatin low,medium and high dose groups was increased,the apoptosis rate in high dose group was higher than that in medium dose group,and that in medium dose group was higher than that in low dose group(all P<0.05).The proliferation and invasion ability of HeLa cells in low,medium and high doses of polydatin were significantly decreased,and the proliferation rate and invasion number of HE-LA cells in high dose group were the lowest(all P<0.05).The expressions of JAK1 and STAT 3 mRNA and protein in HeLa cells of polydatin low,medium and high dose groups were significantly decreased,and the expressions of JAK1 and STAT 3 mRNA and protein in high dose group were lower than those in medium dose group,and those in medium dose group were lower than those in low dose group(all P<0.05).The tumor volume decreased and the tumor quality decreased in the low,medium and high dose groups of polydatin(P<0.05).Conclusion:Polydatin can induce apoptosis of HeLa cells of cervical cancer,inhibit proliferation,migration,invasion and subcutaneous tumor formation in nude mice.The mechanism of action may be related to the regulation of JAK1/STAT 3 pathway signaling pathway.关键词
