Abstract

The difficulty of seed germination of Taxus wallichiana var.chinensis leads to the difficulty of natural regeneration.The dormancy mechanism of T.wallichiana var.chinensis seeds was explored by measuring the water permeability of seed coat,the germination characteristics of embryo and the inhibition effects of the extracts of different seed organs.Through the comparison of germination rate under different seed treatment conditions,the dormancy release technology was explored.The results showed that the seed coat of newly harvested T.wallichiana var.chinensis had good water permeability,and the normal embryo development could meet the needs of seed germination.The presence of endogenous inhibitors was the main reason for seed dormancy.Flowing water washing,hormone soaking,stratification sand storage and other methods could all play roles in relieving the dormancy of the seeds to a certain extent.Among them,the effect of variable temperature stratification on seed dormancy was the most significant.The germina-tion rate of the seeds could reach 58.67%after 180 days of warm temperature stratification and 180 days of low temperature stratification.The effect of hormone soaking was the second.The combination of 200 mg·L-1 GA3 and 10 mg·L-1 6-BA was used for stratification after soaking.The germination rate of the seeds was 22.33%after 6 months of stratification,and the germination rate was 55.00%after 12 months of stratification.The effect of low temperature stratification was better too by washing with running water and then outdoor stratification,and the seed germination rate could reach 43.34%.
