为了探究湖羊钙蛋白酶 3(CAPN3)基因多态性与生长性状的关联性,采用PCR产物直接测序技术筛选湖羊CAPN3 基因单核苷酸多态性(SNPs)位点,进行遗传学分析,并将SNPs与湖羊的生长性状进行关联分析.结果显示:在湖羊CAPN3 基因中检测到 11 个SNPs,其中 3 个为低度多态,8 个为中度多态,有3 个SNPs与湖羊的生长性状显著相关,其中g.42772T>G与湖羊断奶到6 月龄(180 日龄)日增重和6 月龄(181日龄)到周岁日增重显著相关,TT基因型的断奶到6 月龄日增重显著高于GG基因型(P<0.05),TT、TG基因型的 6 月龄到周岁日增重显著高于GG基因型(P<0.05);g.42875C>T与湖羊初生重、断奶到 6 月龄日增重、6 月龄到周岁日增重显著相关,CC基因型的初生重显著高于CT基因型(P<0.05),CC基因型的断奶到 6 月龄日增重显著高于TT基因型(P<0.05),CC与CT基因型的 6 月龄到周岁日增重显著高于TT基因型(P<0.05);g.55993T>C与湖羊胸围显著相关,CC基因型的胸围显著高于TT基因型(P<0.05).提示:这 3 个SNPs与湖羊的生长性状显著相关,可能是湖羊生长性状的重要分子标记.

In order to study the correlation between genetic polymorphisms of calpain 3(CAPN3)of Hu sheep and its growth traits,a di-rect PCR product sequencing technique was used here to screen the sites of single nucleotide polymorphisms(SNPs)of Hu sheep's CAPN3 gene for genetic analysis.Subsequently,a correlation analysis was conducted on SNPs with the growth traits of Hu sheep.The results showed that a total of eleven SNPs were detected in the CAPN3 gene of Hu sheep,and among which there were three SNPs with low polymorphism and eight SNPs with moderate polymorphism.Three SNPs were significantly correlated with the growth traits of Hu sheep.Among them,g.42772T>G was significantly correlated with the daily weight gain of the Hu sheep from its weaning age to the age of six months(180 days of age)and from its age of six months(181 days of age)to the age of one year.The daily weight gain of Hu sheep with TT genotype from the weaning age to the age of six months was significantly hihger than that of the Hu sheep with GG genotype(P<0.05),and the daily weight gain of the Hu sheep with TT and TG genotypes from the age of six months to the age of one year was significantly higher than that of the Hu sheep with GG genotype(P<0.05).g.42875C>T was significantly correlated with the birth weight and daily weight gain of the Hu sheep from the weaning age to the age of six months and from the age of six months to the age of one year.The birth weight of the Hu sheep with CC genotype was significantly higher than that of the Hu sheep with CT genotype(P<0.05),the daily weight gain of the Hu sheep with CC gen-otype from the weaning age to the age of six months was significantly higher than that of the Hu sheep with TT genotype(P<0.05),and the daily weight gain of the Hu sheep with CC and CT genotypes from the age of six months to the age of one year was significantly higher than that of the Hu sheep with TT genotype(P<0.05).g.55993T>C was significantly correlated with the chest circumference of the Hu sheep,with the chest circumference of the Hu sheep with CC genotype significantly higher than that of the Hu sheep with TT genotype(P<0.05).To sum up,there were significant correlations between these three SNPs and the growth traits of Hu sheep,indicating that these SNPs could be used as important molecular markers for the growth traits of Hu sheep.
