旨在观察亚急性瘤胃酸中毒(SARA)前后绵羊瘤胃上皮显微结构与超微结构的变化.选用体况良好的绵羊 15 只,随机分为 2 组,即正常(NG)组(n=5)和SARA组(n=10),其中NG组饲喂精粗比为 3∶7 的全饲料日粮,SARA组饲喂精粗比为 7∶3 的全饲料日粮,SARA模型组诱导成功后,在SARA组中选择5 只绵羊再饲喂精粗比为3∶7 的全饲料日粮,饲喂 25d为恢复(RG)组,动物试验结束后进行屠宰并采集瘤胃组织处腹囊和背囊用于石蜡切片与透射电镜观察形态结构变化.组织学观察结果显示,与NG、RG组相比,SARA组瘤胃乳头部分脱落、变细,形态异常,角质化程度严重,黏膜层变薄,背囊乳头宽度与黏膜上皮厚度均显著低于NG组(P<0.05),而角质层厚度却均显著高于NG组与RG组(P<0.05).透射电镜结果显示,与NG组相比,SARA组瘤胃上皮腹囊与背囊多数棘突层细胞发生坏死,细胞膜破裂导致胞浆内容物丢失,大多数线粒体发生固缩,细胞间桥粒连接较模糊,结构发生损伤,数量减少,细胞器难分辨;与SARA组相比,RG组细胞形态结构正常,线粒体结构完整清晰且数量较多,细胞间桥粒连接模糊,但部分桥粒连接结构完整且数量有所增加.提示:SARA严重影响着瘤胃上皮组织形态结构,可能长期影响瘤胃上皮保护屏障.

SARA(subacute ruminal acidosis)was used to observe the changes in the microstructure and ultrastructure of the rumen epithe-lium in sheep before SARA and after the treatment.Fifteen sheep in good condition were randomly divided into 2 groups:the normal(NG)group(n=5),and the subacute rumen acidosis(SARA)group(n=10).The NG group was fed with a whole-feed diet with a concentrate to forage ratio of 3∶7,and the SARA group was fed with a whole-feed diet with a concentrate to forage ratio of 7∶3.After successful induc-tion of the SARA model group,5 sheep in the SARA group were selected to be fed with a whole feed diet with a concentrate to forage ratio of 3∶7,and feeding for 25 days was defined as the recovery group(RG).At the end of the experiment,the animals were slaughtered,and the ventral and dorsal capsules of the rumen tissue were collected for paraffin section and transmission electron microscope to observe the morpho-logical and structural changes.The HE results showed that,compared with the NG and RG groups,the SARA group had partial exfoliation and thinning of the rumen papilla,abnormal morphology,severe keratinization,and thinning of the mucosal layer.The width of the dorsal capsule papilla and the thickness of the rumen epithelial mucosal layer in the SARA group were significantly lower than those in the NG group(P<0.05).The thickness of stratum corneum was significantly higher than that in the NG and RG groups(P<0.05).The results of trans-mission electron microscopy showed that,in the SARA group,most cells in the spinous process layer of the ventral and dorsal capsules of the rumen epithelium were necrotic,the cell membrane was ruptured and the cytoplasmic contents were lost,most mitochondria were pyknotic,the connections between cells were blurred,the structure was damaged,the number of cells was reduced,and the organelles were difficult to distinguish.Compared with the SARA group,the RG group had normal cell morphology and structure,complete and clear mitochondrial structure and a large number of mitochondria,and fuzzy desmosome connection between cells,but some desmosome connection structure was complete and the number was increased.In conclusion,subacute ruminal acidosis seriously affected the histological morphology of the ru-men epithelium in sheep and might affect the protective barrier of the rumen epithelium in them in the long term.
