猪流行性腹泻病毒流行毒株对4周龄仔猪的致病性研究OA北大核心CSTPCD
Pathogenecity of Porcine Epidemic Diarrhea Virus Field Isolates in 4-Week-Old Piglets
本试验旨在评估猪流行性腹泻病毒(PEDV)流行毒株对4周龄仔猪的致病性.将60头4周龄PEDV抗原抗体阴性断奶仔猪分为4组,每组15头,第1、2、3组试验用猪逐头分别口服2.0mL的PEDV毒株PEDV-FJ06、PEDV-0016和PEDV-TT;第4组作为阴性对照组(NC),逐头口服2.0 mL磷酸盐缓冲溶液(PBS).攻毒后,PEDV-FJ06和PEDV-TT组试验用猪在1 d内即出现水样性腹泻,PEDV-0016组试验用猪在第2 d出现水样性腹泻,各组发病率分别为90%(PEDV-FJ06)、80%(PEDV-0016)、100%(PEDV-TT)和0%(NC).试验用猪在攻毒后第1 d即可通过粪便排毒,排毒高峰期在感染后第2~7 d,至试验结束仍有部分猪持续排毒.攻毒后第3d和第7 d攻毒各组平均日增重均显著低于阴性对照组(P<0.001),试验后期,随着临床症状减轻,被感染猪的健康状况逐步好转,平均日增重也略有恢复,但仍明显低于对照组.攻毒后第7、14和21 d,每组随机选取5头猪剖检,大体病理学检查可见肠腔扩张、肠壁变薄和肠系膜充血等;病变严重的试验用猪组织病理学检查,可见小肠绒毛严重萎缩、脱落,并伴有炎性细胞浸润.免疫组化结果显示,可在感染猪十二指肠、空肠和回肠绒毛上皮细胞胞质中检测到PEDV阳性信号,且攻毒后第7 d攻毒各组试验用猪的回肠组织PEDV免疫组化阳性率为100%.本试验研究表明PEDV-FJ06、PEDV-0016和PEDV-TT毒株对4周龄仔猪均具有较高的致病性,病毒感染后可迅速在小肠上皮细胞中增殖,造成小肠绒毛损伤,进而导致腹泻,并严重影响其平均日增重;PEDV感染后持续排毒,增加了PEDV传播风险.
The study aimed to assess the pathogenesis of porcine epidemic diarrhea virus(PEDV)field isolates in 4-week-old piglets.Sixty PEDV antigen and antibody negative weaned pigs were randomly assigned into four groups at age of 4 weeks,15 piglets each group.Piglets in Groups 1,2 and 3 were orally inoculated with 2.0 mL of field isolates PEDV-FJ06,PEDV-0016 and PEDV-TT,respectively.Piglets in Group 4 were used as a negative control(NC)and orally inoculated with 2.0 mL PBS.Diarrhea was observed in infected piglets beginning at day 1 post inoculation(DPC 1)in Groups 1 and 3,at DPC 2 in Group 2.The morbidity was observed in Groups of PEDV-FJ06(90%),PEDV-0016(80%),PEDV-TT(100%)andNC(0%).PEDV fecal shedding occurred in virus inoculated piglets at DPC 1 and peaked from DPC 2 till DPC 7.Some piglets shed viruses throughout the study.Average daily weight gain(ADWG)in virus inoculated groups was significantly lower(P<0.001)than NC group at DPCs 3 and 7.Five piglets in each group were randomly selected for necropsy at PDCs 7,14 and 21.Gross pathology of virus inoculated pigs included dilation of intestinal bowels,thinning of intestinal walls and mesenteric congestion.Microscopic lesions were atrophic enteritis in small intestines.PEDV specific antigen was detected in villous enterocytes of duodenums,jejunums,and ileums in virus inoculated pigs using immunohistochemistry(IHC),of which 100%positive IHC was observed in ileums at DPC 7.The results from the present study indicated that field isolates PEDV-FJ06,PEDV-0016 and PEDV-TT were all highly pathogenic to 4-week-old piglets and viruses rapidly proliferated in small intestinal epithelial cells,causing damages to intestinal villi,diarrhea and low ADWG.Fecal shedding of PEDV were observed throughout the study,thus increasing the risk of viral transmission.
