Abstract

To investigate the effects of Shengmai Yin with modified ingredients on cardiac function parameters in rat model of exercise-induced fatigue,this study randomly divided 50 healthy male Wistar rats into five groups of 10 rats each.The Control group received 1 mL/(kg·bw·d)of saline orally and was normal fed.The Model group received 1 mL/(kg·bw·d)of saline orally.The Low-,Medium-,and High-dose groups were administered Shengmai Yin with modified ingredients orally at doses of 0.25,0.5,and 1 g/(kg·bw·d),respectively,in equivalent volumes.After completing the 7-week incremental speed treadmill exercise,an exhaustive exercise was conducted.Exhaustion time was recorded,and cardiac blood samples were collected to isolate serum for the determination of myocardial enzyme spectrum,blood gas,and antioxidant indicators.The results showed that rats in the Medium-dose group exhibited significantly longer exhaustion times compared with the Model group(P<0.05).Myocardial enzyme spectrum analysis indicated that compared with the Control group,activities of α-hydroxybutyrate dehydrogenase(α-HBDH),creatine kinase(CK),lactate dehydrogenase(LDH),and aspartate transaminase(AST)were significantly increased in the Model group(P<0.05),with no significant differences observed among the treatment groups(P>0.05).Blood gas analysis revealed that compared with the Control group,the Model group exhibited significantly decreased pH(P<0.05),and significantly increased partial pressure of carbon dioxide(PCO2),bicarbonate(HCO3),anion gap(AnGap),total carbon dioxide(tCO2),and potassium ion(K+)content(P<0.05).Chloride ion(Cl-)and sodium ion(Na+)content significantly increased in both the Model and Treatment groups(P<0.05).Antioxidant indicators analysis indicated that compared with the Control group,superoxide dismutase(SOD)activity and glutathione(GLU)content were significantly decreased(P<0.05),while lactic acid(LD)content was significantly increased(P<0.05)in the Model group.SOD activity was significantly reduced in all Treatment groups(P<0.05),and GLU content was significantly decreased in the Low and Medium-dose groups(P<0.05).Differences in malondialdehyde(MDA)and LD content among the Treatment groups were not significant(P>0.05).These results suggest that administration of Shengmai Yin with modified ingredients can improve exercise performance and prolong exercise time in rats with exercise-induced fatigue.It can reduce enzyme activities in myocardial enzyme spectrum,ameliorate acid-base imbalance and oxidative stress-induced exercise fatigue,thereby exerting anti-fatigue and cardioprotective effects.These findings provide a basis for the development of drugs aimed at alleviating exercise fatigue.
