Effect of Simiao Decoction on Neutrophil Recruitment in Zebrafish with Hyperlipidemia
摘要
Abstract
Objective:To establish the model of zebrafish with hyperlipidemia and investigate the effects of Simiao decoction on lipid metabolism and neutrophil recruitment. Methods:Zebrafish embryos fertilized for 5 days were randomly divided into a blank control group,a model group,high(2 mg/mL),medium(1 mg/mL),and low(0.5 mg/mL)dose groups of Simiao decoction, vitamin D group(10 μmol/L),and atorvastatin calcium group(0.3 μg/mL),with 30 embryos in each group. The hyperlipidemia zebrafish model was constructed by feeding zebrafish with high-fat diet. After 18 days of fertilization, oil red O staining, lipid level, vascular fluorescence imaging and neutrophil recruitment were detected. Results:Compared with model group, zebrafish vascular oil red O dye attachment decreased, and lipid deposition was slightly improved in Simiao decoction low, medium and high dose groups, vitamin D group and atorvastatin calcium group. The levels of TC and TG in the vitamin D group, atorvastatin calcium group,Simiao decoction medium and high dose groups decreased(P<0.05).The vascular fluorescence and wall thickness of the vitamin D group and Simiao decoction high dose group decreased(P<0.05),and the recruitment of neutrophils in the blood vessels was significantly inhibited(P<0.05).Conclusion:Simiao decoction can achieve the treatment of hyperlipidemia by improving the lipid level of hyperlipidemia zebrafish model, reducing the fluorescence intensity and wall thickness of blood vessels, and inhibiting the recruitment of neutrophils in blood vessels.关键词
