目的:探究附子理中汤对非酒精性脂肪肝病(NAFLD)模型大鼠的保护作用及其可能的机制.方法:脂肪乳灌胃建立NAFLD模型大鼠,使用附子理中汤及阳性对照药物易善复治疗.HE和油红O染色观察肝脏和回肠病理损伤和脂肪沉积,全自动生化仪检测血清LDL-C、HDL-C、AST和ALT水平,qRT-PCR检测回肠组织的TNF-α和NF-κB p65 基因水平,ELISA法检测血清TNF-α水平.结果:与对照组相比,模型组大鼠肝脏和回肠发生病理损伤和脂肪沉积,LDL-C、AST、ALT、TNF-α和NF-κB p65 水平均明显上升(P<0.05),HDL-C水平显著下降(P<0.05);与模型组相比,易善复和附子理中汤组大鼠肝脏和回肠组织的脂肪沉积减少,LDL-C、AST、ALT、TNF-α和NF-κBp65 水平均显著下降(P<0.05),HDL-C水平显著上升(P<0.05).结论:附子理中汤可通过抑制TNF-α/NF-κB信号通路减轻NAFLD大鼠肝脏和回肠组织的病理损伤和脂肪沉积.

Objective:To explore the protective effect of Fuzi Lizhong decoction(FLD)on non-alcoholic fatty liver disease(NAFLD)model rats and its possible mechanism.Methods:NAFLD model rats were established by gavage with fat milk,and treated with FLD and positive control drug polyene phosphatidylcholine capsule.HE and oil red O staining were used to observe the pathological damage and fat deposition of liver and ileum.The levels of serum LDL-C,HDL-C,AST and ALT were detected by automatic biochemical analyzer.The levels of TNF-α and NF-κBp65 genes in ileum tissue were detected by qRT-PCR,and the levels of serum TNF-α protein were detected by ELISA.Results:Compared with the control group,the liver and ileum of the model group suffered from pathological damage and fat deposition,and the levels of LDL-C,AST,ALT,TNF-α and NF-κBp65 all increased significantly(P<0.05),while the level of HDL-C decreased significantly(P<0.05).Compared with the model group,the fat deposition in liver and ileum of rats in polyene phosphatidylcholine capsule and FLD groups decreased,and the levels of LDL-C,AST,ALT,TNF-α and NF-κBp65 decreased significantly(P<0.05),while the level of HDL-C increased significantly(P<0.05).Conclusion:Fuzi Lizhong decoction can reduce the pathological damage and fat deposition of liver and ileum in NAFLD rats by inhibiting TNF-α/NF-κB signaling pathway.
