祛毒汤的抗菌抗炎活性及其作用机制研究OACSTPCD
Study on the Antimicrobial and Anti-Inflammatory Activities of Qudu Decoction and Its Mechanism of Action
目的:探索祛毒汤对临床病原菌的抑菌抗炎效果,为中药抗感染提供实验室用药依据.方法:选用多种标准菌株、质控菌株以及临床常见病原菌,用牛津杯扩散法和琼脂平板稀释法测定祛毒汤对菌株的抑菌圈、最小抑菌浓度;菌株生长速率和琼脂平板菌落计数用于检测祛毒汤熏蒸的抑菌和杀菌效果.构建LPS和菌体裂解液刺激的RAW264.7 炎症细胞模型.采用噻唑蓝(MTT)法检测细胞活性;通过检测细胞培养上清液中一氧化氮(NO)、肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)的浓度,评估祛毒汤的抗炎效果.结果:1.祛毒汤对念珠菌具有强力的抗菌效果,对各类细菌也具有一定程度的抗菌效果,存在菌株间的差异性;2.祛毒汤对念珠菌的抗菌活性与现有抗真菌药物的耐药性没有显著相关性,而祛毒汤的抗细菌效果与特殊的耐药细菌存在一定的相关性;3.祛毒汤能有效抑制LPS和菌体裂解液诱导的RAW264.7 细胞NO、TNF-α、IL-6 分泌.结论:祛毒汤具有特定的抗菌谱,对念珠菌最有效;祛毒汤的多药成分的抗真菌机制可能与现有抗生素的耐药机制并不相同,其抗炎机制可能与抑制TNF-α和IL-6 的分泌降低NO的生成有关.
Objective:To explore the antibacterial and anti-inflammatory effects of Qudu decoction against clinical pathogens,providing an experimental basis for its use in treating infections with traditional Chinese medicine.Methods:Various standard strains,quality control strains,and common clinical pathogens were selected to assess the antimicrobial activities of Qudu decoction using the Oxford cup diffusion method and agar dilution method,determining the zones of inhibition and minimum inhibitory concentrations.The growth rates of strains and agar plate colony counts were used to evaluate the bacteriostatic and bactericidal effects of Qudu decoction fumigation.An inflammatory cell model of RAW264.7 was established using LPS and bacterial lysate.Cell viability was assessed by the MTT assay,and the anti-inflammatory effects were evaluated by measuring the concentrations of nitric oxide(NO),tumor necrosis factor-alpha(TNF-α),and interleukin-6(IL-6)in the cell culture supernatants.Results:1.Qudu decoction exhibited strong antimicrobial activity against Candida and showed varying degrees of antibacterial effects against different bacterial strains.2.The antifungal activity of Qudu decoction against Candida was not significantly related to the resistance profiles of existing antifungal drugs,while its antibacterial effects correlated with specific resistant bacteria.3.Qudu decoction effectively inhibited the secretion of NO,TNF-α,and IL-6 in RAW264.7 cells induced by LPS and bacterial lysate.Conclusion:Qudu decoction possesses a specific antimicrobial spectrum,being most effective against Candida.The mechanism of its antifungal activity may differ from the resistance mechanisms of existing antibiotics,and its anti-inflammatory mechanism might be related to the inhibition of TNF-α and IL-6 secretion and the reduction of NO production.
