目的:制备一种可用于肿瘤内注射的温敏水凝胶.本研究设计以吲哚菁绿(ICG)为光敏剂,泊洛沙姆F127 和F68 为凝胶基质的共载吲哚菁绿与苦参碱(MAT)的温敏水凝胶(MAT-ICG-gel),并对其理化性质进行表征.方法:采用冷溶法,以胶凝温度和胶凝时间为评价指标,通过单因素试验筛选出MAT-ICG-gel的最佳制备工艺,并对其形态、流变学特性、光热转换性能、溶蚀性、释放度及初步稳定性等进行考察.结果:MAT-ICG-gel的最优处方组成:F127∶F68=20∶3.5,MAT∶ICG=10∶1.采用最优处方制备MAT-ICG-gel的胶凝温度为(37.3±0.2)℃,胶凝时间为(181±3.6)s,且在 7d内稳定性良好.MAT-ICG-gel 在 37℃时黏度发生骤增,并于 42℃趋于稳定.在 808 nm 激光照射下,MAT-ICG-gel具有良好的光热转换性能.与37℃相比,MAT-ICG-gel在42℃下的溶蚀速率和释药速率均更为缓慢.结论:MAT-ICG-gel制备工艺简单可行,具有良好的光热转换性能、生物可降解性以及温度依赖的药物缓释特性,可为苦参碱联合光敏剂新剂型的研发提供新的思路和理论依据.

Objective:To prepare a kind of thermosensitive hydrogel that can be used for intratumoral injection.In this study,a thermosensitive hydrogel(MAT-ICG-gel)co-loaded with indocyanine green(ICG)and matrine(MAT)was designed using ICG as photosensitizer and poloxamer F127 and F68 as gel matrix.And the physicochemical properties of MAT-ICG-gel were characterized.Methods:The cold solution method was adopted to select the optimal preparation process of MAT-ICG-gel through single factor experiment,with the gelation temperature and gelation time as the evaluation indexes.The morphology,rheological properties,photothermal conversion properties,dissolution properties,in vitro release and preliminary stability of MAT-ICG-gel prepared by using the optimal preparation technology were investigated.Results:The optimal prescription composition of MAT-ICG-GEL was F127∶F68=20∶3.5,MAT∶ICG=10∶1.The optimal formulation was used to prepare MAT-ICG gel,with a gelation temperature of(37.3±0.2)℃,a gelation time of(181±3.6)s,and good stability within 7 days.The viscosity of MAT-ICG-gel increased sharply from 37℃and became stable at 42℃.MAT-ICG-gel exhibited good photothermal conversion performance under 808 nm of laser irradiation.When compared to 37℃,the dissolution rate and drug release rate of MAT-ICG-gel at 42℃were both more slowly.Conclusion:The preparation process of MAT-ICG-gel is simple and feasible.MAT-ICG-gel has good photothermal conversion performance,biodegradability and temperature-dependent sustained drug release characteristics.This study can provide new ideas and theoretical basis for the development of new dosage forms of matrine combined with photothermal therapy.
