黄花倒水莲对糖尿病肾病大鼠肾脏氧化应激和纤维化的影响
Effect of Polygala fallax Hemsl.on Renal Oxidative Stress and Fibrosis in Diabetic Kidney Disease Rats

Abstract
Objective:This study investigated the effects of Polygala fallax Hemsl.(PFH)on renal oxidative stress and fibrosis in rats with diabetic kidney disease.Methods:Twenty-four SD rats were randomly divided into control group(control group),model group(DKD group),PFH low-dose group(PFH-L group)and PFH high-dose group(PFH-H group).All rats except for the ones in control group,were treated with streptozocin combined with high-fat and high-sugar diet to establish diabetic nephropathy rat model.PFH-L group and PFH-H group were gavaged 50 mg/kg and 200 mg/kg extract of PFH,respectively,once a day for 8 weeks.The levels of body mass,blood glucose,24h urinary protein(24h-UP),urea nitrogen(BUN)and blood creatinine(Scr)were measured in each group.H&E staining was used to observe the pathological changes of renal tissue.ELISA was performed to detect the levels of MDA,SOD and GSH-Px in kidney tissues.Masson staining was performed to observe collagen deposition in kidney tissues.The expression levels of fibronectin and collagen Ⅳ were detected by immunohistochemistry.Results:Compared with model group,body mass increased(P<0.05),blood glucose level decreased(P<0.001),24h-UP excretion decreased(P<0.05,P<0.001),BUN level decreased(P<0.05,P<0.001),and Scr level decreased(P<0.05)in the PFH group.The pathological changes of renal tissue were significantly reduced.MDA level in renal tissue decreased(P<0.001),SOD activity increased(P<0.01,P<0.001)and GSH-Px activity increased(P<0.05,P<0.001).Collagen deposition was significantly reduced,and positive expression levels of fibronectin and collagen Ⅳ were significantly reduced(P<0.05 or P<0.001).Conclusion:PFH can improve the oxidative stress status and fibrotic damage in the kidney tissue of DKD model rats.关键词
