目的:探讨黄花倒水莲(PFH)对糖尿病肾病(DKD)大鼠肾脏氧化应激和纤维化的影响.方法:24只SD大鼠随机分为对照组(Control组)、模型组(DKD组)、PFH低剂量组(PFH-L组)和PFH高剂量组(PFH-H组).除对照组外,其余组大鼠采用链脲霉素联合高脂高糖饲料建立糖尿病肾病大鼠模型.PFH-L组、PFH-H组分别灌胃50、200 mg/kg黄花倒水莲提取物,每天1次,连续8周.测量各组大鼠体质量、血糖、24 h尿蛋白(24h-UP)、尿素氮(BUN)、血肌酐(Scr)水平;H&E染色观察肾脏组织的病变,ELISA检测肾组织内MDA、SOD、GSH-Px水平,Masson染色观察肾组织胶原沉积,免疫组化检测纤连蛋白(Fibronectin)、胶原Ⅳ蛋白(Collagen Ⅳ)的表达.结果:与模型组比较,黄花倒水莲治疗组大鼠体质量增加(P<0.05),血糖水平降低(P<0.001),24h-UP排泄量降低(P<0.05,P<0.001),BUN水平降低(P<0.05,P<0.001)、Scr水平降低(P<0.05);肾组织病变明显减轻;肾组织MDA水平降低(P<0.001),SOD活性升高(P<0.01,P<0.001)和GSH-Px活性升高(P<0.05,P<0.001);胶原沉积显著减少,Fibronectin和Collagen Ⅳ阳性表达水平明显降低(P<0.05,P<0.001).结论:黄花倒水莲可改善DKD模型大鼠肾组织氧化应激状态和纤维化损伤.

Objective:This study investigated the effects of Polygala fallax Hemsl.(PFH)on renal oxidative stress and fibrosis in rats with diabetic kidney disease.Methods:Twenty-four SD rats were randomly divided into control group(control group),model group(DKD group),PFH low-dose group(PFH-L group)and PFH high-dose group(PFH-H group).All rats except for the ones in control group,were treated with streptozocin combined with high-fat and high-sugar diet to establish diabetic nephropathy rat model.PFH-L group and PFH-H group were gavaged 50 mg/kg and 200 mg/kg extract of PFH,respectively,once a day for 8 weeks.The levels of body mass,blood glucose,24h urinary protein(24h-UP),urea nitrogen(BUN)and blood creatinine(Scr)were measured in each group.H&E staining was used to observe the pathological changes of renal tissue.ELISA was performed to detect the levels of MDA,SOD and GSH-Px in kidney tissues.Masson staining was performed to observe collagen deposition in kidney tissues.The expression levels of fibronectin and collagen Ⅳ were detected by immunohistochemistry.Results:Compared with model group,body mass increased(P<0.05),blood glucose level decreased(P<0.001),24h-UP excretion decreased(P<0.05,P<0.001),BUN level decreased(P<0.05,P<0.001),and Scr level decreased(P<0.05)in the PFH group.The pathological changes of renal tissue were significantly reduced.MDA level in renal tissue decreased(P<0.001),SOD activity increased(P<0.01,P<0.001)and GSH-Px activity increased(P<0.05,P<0.001).Collagen deposition was significantly reduced,and positive expression levels of fibronectin and collagen Ⅳ were significantly reduced(P<0.05 or P<0.001).Conclusion:PFH can improve the oxidative stress status and fibrotic damage in the kidney tissue of DKD model rats.
