目的:运用中药现代研究方法探讨中药对药"麻黄-半夏"治疗支气管哮喘的机制.方法:通过中药系统药理数据库与分析平台(TCMSP)、DisGeNET及GeneCards检索"麻黄-半夏"的活性成分及作用靶点和支气管哮喘疾病靶点;利用STRING 11.0数据库获得蛋白相互作用(PPI)网络;通过MetaScape软件进行基因本体论(GO)富集分析和京都基因与基因组百科全书(KEGG)通路富集分析,得到"麻黄-半夏"对支气管哮喘的潜在作用通路;采用AutoDockTools、PyMOL软件将主要的活性成分与核心靶点进行对接.结果:从"麻黄-半夏"中筛选得到35个活性成分,241个靶点,其中涉及支气管哮喘作用靶点110个,最终筛选出9个核心化合物和7个核心靶点.KEGG分析提示,"麻黄-半夏"可能通过磷脂酰肌醇3激酶-蛋白激酶B(PI3K-Akt)、白细胞介素(IL)-17、肿瘤坏死因子(TNF)等信号通路发挥对支气管哮喘的治疗作用,分子对接验证亦显示靶点与成分的结合活性均较好.结论:"麻黄-半夏"治疗支气管哮喘具有"多成分-多靶点-多通路"的作用特点,预测"麻黄-半夏"可通过减少血管内皮增生、抑制平滑肌细胞增殖、减轻炎症反应等治疗支气管哮喘,可为后续深入试验研究及其临床应用提供思路及方法.

Objective:To probe into the mechanism of"Ephedra herba-Pinellia ternata"in the treatment of bronchial asthma based on network pharmacology and molecular docking.Methods:The chemical components and potential drug targets of"Ephedra herba-Pinellia ternata"and bronchial asthma targets were searched from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),DisGeNET and GeneCards database.STRING 11.0 database was used to obtain protein-protein interaction(PPI)network.Gene ontology(GO)analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis were carried out by MetaScape software to obtain the potential pathway of"Ephedra herba-Pinellia ternata"on bronchial asthma.AutoDockTools and PyMOL software were used to dock the main active components with the core targets.Results:Totally 35 active components and 241 targets were extracted from"Ephedra herba-Pinellia ternata",of which 110 targets were related to the action of bronchial asthma,and finally 9 core compounds and 7 core targets were screened out.KEGG enrichment analysis indicated that"Ephedra herba-Pinellia ternata"may acted on bronchial asthma through phosphatidylinositol 3-kinase-protein kinase B(PI3K-Akt),interleukin(IL)-17,tumor necrosis factor(TNF)and other signal pathways.Molecular docking results showed that the binding activity of targets and components were good.Conclusion:The treatment of bronchial asthma with"Ephedra herba-Pinellia ternata"is characterized by"multicomponent-multitarget-multipathway",and it is predicted that the treatment of bronchial asthma with"Ephedra herba-Pinellia ternata"can be achieved by decreasing the endothelial hyperplasia,inhibiting the proliferation of smooth muscle,and reducing the inflammatory response,which can provide ideas and methods for the subsequent in-depth experimental research and clinical application.
