摘要

Abstract

To explore the pathogenesis of breast cancer brain metastasis(BCBM),the biological functions of differentially expressed genes in BCBM were investigated and the hub genes of BCBM were screened in this study.Four gene expression profiles of BCBM datasets(GSE12237,GSE100534,GSE12598,and GSE43837)were retrieved from the gene expression omnibus(GEO)database and screened by R packages to obtain the differentially expressed genes of BCBM.The functional annotations of differentially expressed genes were conducted by gene ontology(GO)analysis.The enrichment analysis of signal pathways of differentially expressed genes was explored by Kyoto encyclopedia of genes and genomes(KEGG)analy-sis.The protein-protein interaction network was structured by the STRING database to screen the hub genes that might par-ticipate in BCBM.Finally,survival analysis was conducted by Kaplan-Meier survival analysis database.As a result,a total of 261 differentially expressed genes present in 2 or more datasets were screened out.Functional analysis showed that those differentially expressed genes were related to biological processes such as extracellular matrix organization and extracellular structural organization,molecular functions such as extracellular matrix structural composition and collagen binding,and cellular components such as collagen-containing extracellular matrix and collagen trimer.Signal pathways were mainly en-riched in protein digestion and absorption,focal adhesion pathways.Nine hub genes were screened by using protein interac-tion network analysis,among which two hub genes(DCN,COL6A1)were significantly correlated with the survival proba-bility of BCBM.The two hub genes could serve as potential hub genes for BCBM and might provide ideas for molecular mechanism research.
