Abstract

Streptococcus suis(S.suis)is one of the most important zoonotic pathogens and brings great losses to the pig industry.The objective of the present study was to evaluate the immunoprotective effect of the recombinant phosphate ABC transporter ATPase of S.suis in mice.The vector for the phosphate ABC transporter ATPase of S.suis was constructed and the recombinant ATPase protein was expressed via prokaryotic expression.The resulting recombinant ATPase protein was used to prepare a subunit vaccine for mouse immunization.The ELISA titers of serum antibodies,immunoprotection rate,bacterial loads in different organs and inflammatory factors were examined at two weeks after a booster immunization.The results showed that the recombinant protein mainly existed in supernatant of E.coli culture.The recombinant protein induced a high level of IgG antibodies and 60%immunoprotection against S.suis serotype 2 infection in mice.In addition,bacterial loads were significantly reduced in brains,hearts and lungs of the immunized mice and the mRNA expression levels of IL-1β,IFN-γ,IL-6,IL-8 and TNF-α were also significantly up-regulated.The results from this study indicated that recombinant phosphate ABC transporter ATPase was a potential subunit vaccine candidate for prevention of S.suis serotype 2 infection.
