Mitochondrial genomic characterization and phylogenetic analysis of Perilla citriodora(Lamiaceae)

Abstract
[Purpose]This study aimed to enrich the genomic data of Perilla citriodora by annotating and assembling its mitochon-drial genome,thereby providing a reference for germplasm evaluation and the breeding of new varieties of this diploid species.[Method]The entire mitochondrial genome of Perilla citriodora was sequenced,annotated,and assembled using a combination of Illumina and PacBio datasets.[Result]The assembled mitochondrial genome was 301 156 bp long,with a GC content of 45.23％,and exhibited a typical circular structure.In total,59 unique genes were identified,including 37 protein-coding genes,20 tRNA genes,and 2 rRNA genes.Among the protein-coding genes,eight contained 18 introns.The mitochondrial genome indicated a high preference for A/T codon usage.Furthermore,180 dispersed repeat sequences,78 simple sequence repeats,and 6 tandem repeat sequences were detected.Additionally,70 sequence fragments(12 680 bp)were transferred from the chloroplast to the mitochondri-a,accounting for 4.23％of the entire mitochondrial genome.Phylogenetic analysis revealed that within the Lamiaceae family,Perilla citriodora was most closely related to Salvia miltiorrhiza and Platostoma chinense.Interspecies comparisons indicated that the Ka/Ks ratio of the 28 protein-coding genes was less than 1.[Conclusion]The results of this study enhance the mitochondrial genome data of Perilla citriodora and provide a theoretical basis for germplasm resource evaluation and the breeding of new varieties of this diploid species.关键词
