为探讨丹参对黄瓜花叶病毒(CMV)侵染的生理响应机制,采用人工注射接种方法,设置注射接种CMV的丹参(CMV组)、仅空载体pCB301注射接种的丹参(MOCK组)以及未接种CMV的丹参(CK组)3个处理,比较分析接种CMV前后丹参苗根系生长、植株各部位代谢产物和生理特性的差异.结果表明,CMV侵染能够抑制丹参植株和根系的生长,破坏其内部代谢平衡.与MOCK组相比,CMV组侵染丹参的根长、须根数等各项根系指标均下降,根、茎和叶的可溶性蛋白、游离脂肪酸、总酸、总酚含量上升,总黄酮和可溶性糖含量则有所下降.与MOCK组相比,CMV组侵染丹参的丹酚酸类成分含量升高幅度最高可达35.12%,而丹参酮类成分含量降低幅度最高可达96.65%.CMV组侵染丹参根、茎、叶中的丙二醛(MDA)和过氧化氢(H2O2)含量以及超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性均明显高于MOCK组,最高分别上升78.53%、57.09%、58.95%、38.51%和58.86%.综上,为维持自身稳态,CMV侵染丹参植株生理指标发生明显变化,其中可溶性蛋白、MDA和H2O2含量及SOD和POD活性对CMV侵染的反应较为强烈,可作为丹参抗CMV评价的生理指标.本研究结果为丹参与植物病毒互作机制研究及相关抗病育种工作提供了理论依据.

To investigate the physiological response mechanisms of Salvia miltiorrhiza to cucumber mosaic virus(CMV)infection,an artificial inoculation was employed.Three treatments were set up:S.miltiorrhiza inoculated with CMV(CMV group),S.miltiorrhiza inoculated with the empty vector pCB301 only(MOCK group),and S.miltiorrhiza that was not inoculated(CK group).Comparative analyses were conducted to examine the differences in root growth,metabolite products,and physiological characteristics of S.miltiorrhiza seedlings before and after CMV inoculation.The results indicated that CMV infection could inhibit the growth of S.miltiorrhiza plants and roots,and disrupt their internal metabolic balance.Compared to the MOCK group,the CMV-infected group exhibited a decrease in various root parameters,such as root length and the number of fibrous roots.The content of soluble protein,free fatty acids,total acid,and total phenolics in roots,stems,and leaves were increased,while the content of total flavonoids and soluble sugars were decreased.In the CMV-infected group,the content of salvianolic acids in S.miltiorrhiza was increased by 35.12%compared to the MOCK group,while the content of tanshinones was decreased by 96.65%.Additionally,the levels of MDA and H2O2,as well as the activities of SOD,POD,and CAT in the roots,stems,and leaves of the CMV-infected group were significantly higher than those in the MOCK group,with increases by 78.53%,57.09%,58.95%,38.51%and 58.86%,respectively.In summary,to maintain homeostasis,CMV infection causes significant changes in the physiological indicators of S.miltiorrhiza.Among these changes,the content of soluble protein,MDA and H2O2,as well as the activities of SOD and POD,responed strongly to CMV infection and can be used as physiological indicators for evaluating the resistance of S.miltiorrhiza to CMV.The results of this study provide a theoretical basis for research on the interaction mechanism between S.miltiorrhiza and plant viruses,as well as for related disease-resistant breeding.
