裂片石莼水培液对海水养殖环境分离弧菌的抑制特性及其活性物质类型分析OA北大核心CSTPCD
Inhibitory performance of Ulva fasciata hydroponic solution on Vibro from mariculture and analysis of its active substances
活体大型海藻能有效控制水产养殖环境病原弧菌污染,与其分泌的抑菌活性物质密切相关,然而有关活体大型海藻分泌物质类型及其对海水养殖环境弧菌的抑制特性报道较少.通过制备不同密度(1.25~10 g/L)和培养时间(1~3 d)下的裂片石莼水培液,并从不同海水养殖场筛选分离了 6株耐药弧菌,包括 1 株副溶血弧菌(Vibrio parahaemolyticus,Vp.)、1 株锡那罗州弧菌(Vibrio sinaloensis,Vs.)、1 株梅氏弧菌(Vibrio metschnikovii,Vm.)和 3 株溶藻弧菌(Vibrio alginolyticus,Va.),分析不同裂片石莼(Ulva fasciata)水培液的结构组成及其对不同耐药弧菌的抑制特性.结果表明,裂片石莼水培液与弧菌共培养12h时,各弧菌受到不同程度的抑制,且在10 g/L抑制效果较好;Vp.W1、Vs.W2和Vm.R3在培养3 d(64%~100%)的裂片石莼水培液作用下抑制效果显著高于第1 d(1.32%~39.42%);溶藻弧菌在培养1 d和3 d的裂片石莼水培液作用下抑制效果差异不显著,抑制率均不超过35%.SPE-GC-MS分析发现,裂片石莼水培液含有丰富的活性物质,包括酸类、醇酚类、醛酮类、烷烃和芳香烃、杂环、酯类等多种化合物,且以邻苯二甲酸单(2-乙基己基)酯、乙缩醛、2-(4-羟基苯)乙醇和2,4,6-三溴苯酚等物质为主.
Live macroalgae serve as an effective strategy to control pathogenic Vibrio(V.)pollution in mariculture,primarily due to the antibacterial active substances they secrete.However,limited information is available on the types of substances secreted by live macroalgae and their inhibitory performance on Vibrio in the mariculture environment.In this study,hydroponic solutions of Ulva fasciata(HSUF)were prepared at varying densities(1.25 g·L-1-10 g·L-1)and culture times(1-3 d).Six antibiotic-resistant Vibrio strains were isolated from the mariculture environment,including Vibrio parahaemolyticus(Vp.W1),Vibrio sinaloensis(Vs.W2),Vibrio metschnikovii(Vm.R3),and Vibrio alginolyticus(Va.W1,Va.S1,Va.M3).The structural composition of HSUF and their inhibitory performance against the isolated Vibrio strains were further analyzed.With the 12 h-culture of 3-d HSUF at different densities,the inhibi-tion rates were superior at 10 g·L-1.The inhibition rates of Vp.W1,Vs.W2,and Vm.R3 were significantly higher with 3-d HSUF(64%-100%)than with 1-d HSUF(1.32%-39.42%).However,there was no significant difference in inhibi-tion of Vibrio alginolyticus strains between 1-d and 3-d culture HSUF,with all inhibition rates being lower than 35%.Solid phase extraction-gas chromatography-mass spectrometry analysis revealed that HSUF contained a rich array of active substances,including acids,alcohol phenols,aldehydes and ketones,alkanes and aromatic hydrocarbons,het-erocycles,and esters.Moreover,four active substances(2,4,6-tribromophenol,2-(4-hydroxyphenyl)ethanol,acetal,and mono(2-Ethylhexyl)phthalate)were primarily found in HSUF.
