目的 观察葛根素对高糖条件下大鼠足细胞沉默信息调控因子1(SIRT1)/叉头转录因子O3(FOXO3)自噬通路的影响.方法 在高糖条件下,采用蛋白免疫印记、免疫荧光等方法检测足细胞自噬相关蛋白 SIRT1、FOXO3a、p62 和Beclin-1 的表达.结果 HG组足细胞Beclin-1 表达较NG组下调,足细胞p62 的表达较NG组上调,SIRT1/FOXO3 自噬通路受到抑制,而应用葛根素后可使Beclin-1 表达上调,p62 的表达下调,SIRT1/FOXO3 自噬通路恢复,差异有统计学意义(P<0.05,P<0.01).结论 葛根素可逆转高糖条件下足细胞自噬的抑制作用,有望成为一种治疗糖尿病肾病的新型药物.

Objective To observe the effects of puerarin on the autophagy pathway of silencing information regulator 1(SIRT1)/forkhead transcription factor O3(FOXO3)in rat podocytes under the condition of high glucose,and to explore the therapeutic effect of puerarin on diabetic kidney disease(DKD).Methods The expressions of autophagy-related proteins SIRT1,FOXO3a,p62 and Beclin-1 were detected by protein immunoblotting and immunofluorescence under high glucose con-dition.Results Beclin-1 expression in rat podocytes of high glucose group was down-regulated,p62 expression in podocytes was up-regulated,as compared with NG group,and SIRT1/FOXO3 autophagy pathway was inhibited.However,after the ap-plication of puerarin,Beclin-1 expression was up-regulated,p62 expression was down-regulated,and SIRT1/FOXO3 autoph-agy pathway was restored,showing significant difference(P<0.05,P<0.01).Conclusion Puerarin can reverse the inhi-bition of podocyte autophagy under high glucose condition,and can be expected to be a new drug for the treatment of DKD.
