哮喘对膝骨关节炎影响的孟德尔随机化研究
Mendelian randomization study on effects of asthma on knee osteoarthritis

Abstract
Objective To investigate the effects of asthma on knee osteoarthritis(KOA)by means of Mendelian ran-domization(MR)method.Methods Inverse variance weighting(IVW)was used as the main evaluation method.At the same time,four methods were used to test the reliability and stability of the results:MR Egger regression,weighted median(WM),simple estimation based on mode,and weighted estimation based on mode.Leave-one-out method was used for sensi-tivity analysis of research results.Cochran's Q-test was used to calculate intercept and P value,and analyze the heterogeneity of SNP of IVW and MR-Egger results.The MR-pleiotropy function was used to analyze the research results.Two sample MR as-sessments were conducted to assess the causal relationship between asthma(main datasets and secondary datasets)and KOA,and reverse MR were conducted to investigate the effects of KOA on asthma.Results The asthma main dataset was associated with an increased probability of KOA occurrence(OR=1.664,95％CI:1.134-2.441,P=0.009).The asthma secondary datasets were associated with an increased probability of KOA occurrence(OR=6 405.249,95％CI:1.850-22 176 818.850,P=0.035).The results of leave-one-out method showed that the positive MR result was stable.Reverse MR studies showed that KOA was not associated with an increased probability of asthma occurrence(P＞0.05).Conclusion Asthma can pro-mote the occurrence and development of KOA to a certain extent,and is a potential adverse factor for KOA.关键词
