波棱瓜子总木脂素的急性和亚慢性毒性试验OA北大核心CSTPCD

旨在用小鼠急性毒性试验和大鼠亚慢性毒性试验评价波棱瓜子总木脂素的安全性.在 24 只ICR小鼠急性经口毒性试验中,设单次最大给药量(5 000 mg/kg),给药后连续观察7d;在亚慢性毒性试验中,40 只SD大鼠分为对照组(0.5%羧甲基纤维素钠)和波棱瓜子总木脂素低(100 mg/kg)、中(200 mg/kg)和高(400 mg/kg)剂量组进行灌胃给药,每天1 次,连续给药 30 d,灌胃结束后测定大鼠体重、采食量、血液指标、生化指标、脏器系数和病理组织学变化.结果:在小鼠急性毒性试验中,波棱瓜子总木脂素对ICR小鼠的LD50>5 000 mg/kg,按照外源化合物急性毒性分级标准波棱瓜子总木脂素属于实际无毒;大鼠亚慢性毒性试验中,与对照组比较,波棱瓜子总木脂素各剂量组体重、采食量、血液学、血液生化学、脏器系数均无显著差异(P>0.05),组织病理学检查结果未见与波棱瓜子总木脂素毒性有关的病理变化.综上,波棱瓜子总木脂素临床合理剂量使用安全性高.

In order to evaluate the safety of total lignans of Semen herpetospermi,the acute toxicity and subchronic toxicity tests were car-ried out in mice and rats,respectively.In the acute transoral toxicity test,observation lasted for 7 days after the administration of a maximum single dose(5 000 mg/kg).In the subchronic toxicity test,40 SD rats were randomly divided into the control group(0.5%sodium carboxy-methyl cellulose),the low(100 mg/kg),medium(200 mg/kg)and high(400 mg/kg)dose groups.The rats were administered by gavage once a day for 35 consecutive days.At the end of the experiment,the body weight,food consumption,blood index,biochemical index,organ coefficient and histopathological changes in the rats were determined.The results showed that,in the acute toxicity test,the LD50 of total lig-nan of Semen herpetospermi in the ICR mice was higher than 5 000 mg/kg.It was suggested that the total lignans was actually non-toxic ac-cording to the classification standard of acute toxicity of exogenous chemicals.For the subchronic toxicity test,there were no significant differ-ences in hematology,coagulation indicators,blood biochemistry,and organ coefficients among the total lignans groups and the control group(P>0.05).The histopathological examination revealed that there were no significant pathological changes related to the total lignans.These results indicated that total lignans of Semen herpetospermi could be used in veterinary clinic practice at an appropriate dose.
