Abstract

Objective To screen key genes in the occurrence and development of colorectal cancer and analyze their relation-ship with survival prognosis and immune infiltration.Methods The gene expression profile of colorectal cancer GSE178145 was downloaded from Gene Expression Omnibus(GEO).Differentially expressed genes(DEGs)between colorectal cancer and normal tissues were screened using R software.The biological effects of these genes were determined by gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG).A protein-protein interaction(PPI)network was con-structed to identify the key genes in the progression of colorectal cancer,and their expressions and survival prognosis were verified using the GEPIA2 database.Finally,the TIMER database was employed to analyze the correlation between progno-sis-related genes and immune infiltrating cells.Results A total of 897 DEGs were screened.GO analysis revealed that the DEGs mainly focused on the regulation of metal ion transport,ion channel activity,and fiber shrinkage.KEGG pathway a-nalysis indicated that the DEGs were mainly involved in cytokine-receptor interaction and calcium signal transduction.Ten key genes were obtained by PPI analysis,and it was found from the GEPIA2 database that CXCL2,IL-1β,CXCL10,and UPS18 showed consistent and significant differences.Survival analysis found that CXCL2 and IL-1β prolonged the survival rate of patients with colorectal cancer.Immune infiltration analysis showed that the expressions of CXCL2 and IL-1β were related to macrophage infiltration.Conclusion CXCL2 and IL-1β are key genes in the occurrence and development of color-ectal cancer,and are related to the survival,prognosis,and immune infiltration of colorectal cancer.
