多房棘球蚴感染小鼠肝转录组分析及肝纤维化关键基因的筛选与验证OA北大核心CSTPCD
Transcriptome analysis and critical gene of fibrosis screening and verification of mice liver infected with Echinococcus multilocularis larvae
为探究多房棘球蚴感染引起宿主肝纤维化的关键基因变化,通过建立多房棘球蚴感染小鼠模型,利用高通量测序技术获得感染小鼠肝不同时间点差异mRNA表达谱,结合DisGeNET、GeneGards等数据库中已收录的肝纤维化相关基因集,挖掘感染进程中影响肝纤维化的重要基因.结果显示,多房棘球蚴感染小鼠肝出现胶原纤维沉积;通过转录组测序数据结合vlookup函数检索,发现2 943个肝纤维化相关基因在感染后不同时间出现差异表达,其中显著上调的基因主要富集在PI3K-Akt等通路上,实时荧光定量PCR验证结果与高通量测序结果一致,通路中神经生长因子(Ngf)、肌腱蛋白C(Tnc)基因在肝星状细胞中表达量显著高于肝实质细胞、肝窦内皮细胞和枯否细胞,在小鼠原代肝星状细胞中敲低Ngf基因影响胶原基因Col1a1及Timp1的表达,推测这些通路和基因可能参与多房棘球蚴感染后致小鼠肝组织纤维化的过程.
In order to explore the key gene changes in host liver fibrosis caused by Echinococcus multilocularis larvae infection,a mouse model of E.multilocularis larvae infection was established,and high-throughput sequencing technology was utilized to obtain mRNA expression profiles of the liver at different time points following infection with E.multilocularis larvae.Combined with DisGeNET,GeneGards and other databases,gene sets related to liver fibrosis were minded to explore the important genes affecting liver fibrosis during infection.The results showed that collagen fiber deposition occurred in the livers of mice infected with E.multilocularis larvae.Transcriptome sequencing,combined with the vlookup function for data retrieval,revealed that 2 943 liver fibrosis-related genes were differentially expressed at different times during the E.multilocularis larvae infection.Among them,genes showing significant up-regulation were predominantly enriched in PI3K-Akt and other pathways.The results of real-time fluorescence quantitative PCR were consistent with those of high-throughput sequencing.In the pathway,the expression levels of nerve growth factor(Ngf)and Tenascin C(Tnc)genes were higher in hepatic stellate cells compared to hepatic parenchymal cells,hepatic sinusoidal endothelial cells and Kupffer cells.Knockdown of Ngf gene in mouse primary hepatic stellate cells affected the expression of collagen genes Col1α1 and Timp1,indicating that these pathways and genes might play roles in the development of liver fibrosis in mice following E.multilocularis infection.
